Abstract: These data suggest that AD may play an anti-inflammatory role in the pathophysiology of RA.

“…On the other hand, the high adiponectin levels of patients with RA can also be interpreted as an attempt to overcome the well-known pro-inflammatory effect of leptin, for example by counteracting the proinflammatory effects of TNF-a and reducing the production of IL 6 and C reactive protein in RA. In keeping with this data, recent reports showed that adiponectin mitigates the severity of collagen-induced arthritis in mice (Lee et al 2008) whereas another work showed that Etanercept (a soluble fusion protein that specifically binds to TNF-a) treatment in RA patients is able to increase adiponectin levels in women with RA (Lewicki et al 2008). This finding has been also suggested by another author using another anti TNF-a treatment such as infliximab (Nagashima et al 2008).…”

“…In contrast, Lee et al observed, that adiponectin significantly mitigated the severity of arthritis in collagen-induced arthritis in mice, with decreased expression of TNF-α, IL-1β, MMP-3 in RA synovial fibroblasts. These data suggest an anti-inflammatory adiponectin role in the pathophysiology of RA [38]. 10 In clinical observations, it was reported, that adiponectin plasma levels of RA pts did not differ significantly [21] or were markedly increased in comparison with healthy controls [21,27].…”

“…In contrast to the results in vitro, in the primarily inflammation-driven arthritis model of collagen-induced arthritis (CIA), gene transfer or intraarticular injection of adiponectin resulted in a reduction in the severity of arthritis in mice with CIA (37,38). When the adenoviral vector was applied prior to progression of arthritis, the histology scores for inflammation and joint erosion were significantly decreased without alteration of the anticollagen levels (37).…”
