Adiponectin and leptin exert antagonizing effects on proliferation and motility of papillary thyroid cancer cell lines

Abstract: Adiponectin (Acrp30) and leptin, adipokines produced and secreted mainly by the adipose tissue, are involved in human carcinogenesis. Thyroid carcinomas are frequent endocrine cancers, and several evidences suggest that they are correlated with obesity. In this study, we first analyzed the expression levels and prognostic values of Acrp30, leptin, and their receptors in thyroid cancer cells. Then, we investigated the role of Acrp30 and leptin in proliferation, migration, and invasion. We found that Acrp30 trea… Show more

“… 218 , 219 Secondly, inflammatory adipokines secreted by WAT, such as IL-6, leptin, and adiponectin, can influence STAT3, AKT, JNK, and MEK/ERK signal pathways in cancer cells directly, consequently upregulating the expression of proliferation and invasion-related genes. 220 222 Finally, adipocytes are also involved in building a beneficial microenvironment for cancer cells. For example, adipocytes can decrease the natural killer cell toxicity, activate the inflammatory phenotype of macrophages, and promote fibrosis and vascularization in tumors.…”

“…This was demonstrated in leptin expression where it decreased in D7 cells but was not significantly reduced in D5 cultures. The effect of adiponectin on leptin expression is remarkably limited, and instead, adiponectin appears to influence leptin-mediated affects by antagonizing leptin activity [54,55], in part by abrogating the signal transduction capacity of its receptor [56][57][58]. Nevertheless, Jarde et al [59] reported a tendency towards a reduction in leptin expression in adiponectin-treated MCF-7 breast cancer cells.…”
