Adding to Yersinia enterocolitica Gene Pool Diversity: Two Cryptic Plasmids from a Biotype 1A Isolate

Abstract: We report the nucleotide sequence of two novel cryptic plasmids (4357 and 14 662 base pairs) carried by a Yersinia enterocolitica biotype 1A strain isolated from pork. As distinguished from most biotype 1A strains, this isolate, designated 07-04449, exhibited adherence to eukaryotic cells. The smaller plasmid pYe4449-1 carries five attributable open reading frames (ORFs) encoding the first CcdA/CcdB-like antitoxin/toxin system described for a Yersinia plasmid, a RepA-like replication initiation protein, and mo… Show more

“…Two cryptic plasmids, pYe4449-1 and pYe4449-2, described in another BT 1A strain (Lepka, et al, 2009) show no similarity to the plasmids described here apart from the mobilization region mobABC. These plasmids add to the overall gene pool in Y. enterocolitica, especially the non-pathogenic BT, but their function and impact is unknown.…”

“…This is important, as pYV itself is non-conjugative. Interestingly, pYE854 was isolated from a non-pathogenic BT 1A strain, which is known never to carry the virulence plasmid (Cornelis, et al, 1987;Lepka, et al, 2009;Wren 2003). The study suggests though that self-transmissible plasmids are common amongst Yersiniae as they also found a plasmid related to pYE854 which could also mobilize pYV (Hammerl, et al, 2008).…”

“…This is not unexpected, given the extraordinary genetic diversity of the species. 2,36 However, novel strategies including shotgun proteomics and de novo MS/MS spectra interpretation 37 may help to characterize the secretome of Y. enterocolitica biotype 1A strains in the near future.…”
