Activity of ectonucleotidases and adenosine deaminase in rats exposed to cigarette smoke

Abstract: Cigarette smoke is a complex mixture of various toxic substances that are capable of initiating oxidative damage and promoting blood platelet alterations. In this study, we investigated the activities of the ectoenzymes NTPDase (ectonucleoside triphosphate diphosphohydrolase, CD39) and 5'-nucleotidase (CD73) in platelets as well as adenosine deaminase (ADA) in the plasma of rats exposed to aged and diluted sidestream smoke during 4 weeks. The rats were divided into two groups: I (control) and II (exposed to sm… Show more

“…Surprisingly, we found an increase in CD39 expression of inflammatory cells and soluble ATPase activity in the airways of smokers and patients with COPD. Similar observations were also reported in platelets after acute smoke exposure [22,23]. However, in platelets an opposing effect of acute cigarette smoke stimulation on ATP hydrolysing capacity has also been noted [24].…”

“…Furthermore, observed an increase in the activity of ADA in the cerebral cortex of rats eight days after neonatal Hypoxia Ischemia [28] and the administration of Metothrexate (MTX) induced a significant decreased in the ADA activity in the cerebral cortex, kidney and liver tissues of young rats [29]. Besides, ADA activity is decreased in plasma of rats exposed to aged and diluted sidestream smoke [30], and in lymphocytes and platelets of patients with multiple sclerosis [31,32].…”

“…Control animals were placed in an equal chamber for the same amount of time but without exposure to smoke. When the smoke exposure was carried out, the control group was always outside, without any contact with smoke …”

“…A metal grille was placed on top of the cigarette holder to avoid direct contact with the cigarettes and, thus, prevent the rats from injuring themselves. The inhalation exposure of our study was to aged and diluted sidestream smoke, used as a surrogate of environmental tobacco smoke as experienced by non‐smokers …”
