Objective The etiology of oral candidiasis (OC) was Candida albicans, C. krusei, C. dubliniensis, C. tropicalis that are frequently found in human immunodeficiency virus/ acquired immunodeficiency syndrome (HIV/AIDS) patients. Marine ascomycetes (MA) have been widely reported as an important producer of various antibiotic compounds. However, there is limited study of antifungal compounds from MA against Candida species. The aim of this study was to investigate the antifungal susceptibility of MA against Candida spp. isolates from OC HIV/AIDS patient. Materials and Methods Trichoderma sp. is a sponge-associated fungus collected from Karimunjawa National Park, Central Java, Indonesia. The validation of C. albicans, C. krusei, C. dubliniensis, C. tropicalis. was done by ChromAgar. This study was true experimental with post-test only control group design; the sample was four replications for each group. Nystatin administration (K +), the golden standard antifungal drug, was used. The minimum fungicidal concentration (MFC), minimum inhibitory concentration (MIC), and diffusion zone methods were done. Analysis of variance difference test, and post-hoc Tukey's honest significant different were done to analyze the significant different between groups (p ≤ 0.05). Results The MFC and MIC of MA against C. albicans, C. krusei, C. dubliniensis, and C. tropicalis were found at 12.5%. In addition, the greatest diffusion zone of MA against C. albicans, C. krusei, C. dubliniensis, and C. tropicalis was found at 12.5%. There is no appreciable difference in antifungal activity between K + and 12.5% of MA extract (p ≥ 0.05). Conclusion Concentration of 12.5% MA extract has antifungal susceptibility against Candida spp. isolates from OC HIV/AIDS patient.

Methicillin-Resistant Staphylococcus aureus (MRSA) is a species of S. Aureus, which shows resistance to the methicillin class of antibiotics. MRSA infection poses significant health problems because it causes severe disease in a vulnerable population and also because effective antibiotics are limited. Therefore, the research to find the new anti-MRSA compound is a necessity. The genome mining approach through Biosynthetic Gene Clusters (BGC) analysis can detect the ability of microbes to produce antibiotics. Thus, the study aimed to screen potential anti-MRSA microbes and predict their BGC. This study used ten microorganisms isolated from mangrove areas in Segara Anakan Cilacap and clinical MRSA 2983 from the Faculty of Medicine, UNSOED. All samples microorganisms were cultivated in starch casein nitrate agar. DNA isolation used Quick-DNA Fungal/Bacterial Miniprep Kit, 16S rRNA gene amplification used 27F and 1492R primers, and DNA amplicons were sequenced by the Sanger sequencing method. BCG prediction of the associated genes used antiSMASH 5.1.2. The results showed that W-5A, P-6B, and W-5B isolates have anti-MRSA activities with inhibition indexes of 0.58; 0.53; and 0.47, respectively. Species identification revealed similarity of W-5A with Streptomyces longisporoflavus strain Moghannam M1, P-6B with Ochrobactrum intermedium LMG 3301, and W-5B with Streptomyces cellulosae strain NBRC 13027. The results of BGC analysis showed that Streptomyces longisporoflavus has one region involved in the synthesis of tetronasin. Ochrobactrum intermedium has one region involved in the synthesis ambactin. Streptomyces cellulosae has eight gene clusters for tiacumicin B, actinorhodin, ulleungmycin, albaflavenone, desferrioxamine B/E, stenothricin, auricin, and prejadomycin productions. These results indicate that the microbes isolated from Segara Anakan Cilacap serve as promising antibacterial producers.

Utilization of winged bean (Psophocarpus tetragonolobus) seed powder as a coagulant and activated natural zeolite as an adsorbent for improving of tapioca and tofu wastewater qualities

Water treatment of tapioka and tofu wastewater was studied. Water treatment was conducted by coagulation process by winged bean (Psophocarpus tetragonolobus) seed powder which was followed by adsorbstion by natural active zeolite. The changing of turbidity, DO and pH during water treatment were investigated. Turbidity of tapioca wastewater which was initially 798 FNU, decreased to 57 FNU. Increasing of tapioca wastewater DO value was obtained from 1.20 to 5.92 ppm. Increasing pH tapioca wastewater was also observed from 5.8 to 6.7. Similar results were obtained for tofu wastewater. Turbidity of tofu wastewater which was initially 680 FNU, decreased to 37 FNU. Increasing of tofu wastewater DO value was obtained from 1.67 to 6.61 ppm. Increasing pH tofu wastewater was also observed from 4.7 to 6.7. Results showed that the improvement qualities of tapioca and tofu wastewater was achieved which meet wastewater quality standard by Minister of Environment of the Republic of Indonesia.
