Activity Measurement of Chitosanase by an Amperometric Biosensor

Abstract: An amperometric biosensor for the detection of glucosamine (GlcN) and chitosan oligosaccharides ((GlcN)n) was introduced for an activity measurement of chitosanase. By using the biosensor, an increase in the anodic current due to the production of GlcN and (GlcN)n by chitosanase was measured in a chitosan solution. The maximum value of the slope of the current increase was proportional to the enzyme concentration up to 1.4 mg mL -1 . The present method had the advantages of being simple and rapid over the conv… Show more

“…The reaction mixture, consisting of 0.4 ml of 1.0% substrate in PBS and 0.1 ml of enzyme solution in 1% BSA-PBS, was incubated for 10-120 min at 37 ° C, and the amino sugars released were determined [Blix, 1948;Tatsumi et al, 2009]. One unit of activity was defined as the amount of enzyme catalyzing the production of 1 mol of the amino sugar per min, using glucosamine as the standard.…”

“…However, there are a few methods to determine glucosamine in different matrixes based on its characteristics as hexosamine, such as Elson-Morgan colorimetric method [2,3]. Other analytical instrumental methods to determine amino sugars have been also implemented: radioisotope dilution [4], capillary electrophoresis [5], amperometric biosensor [6], and high performance liquid chromatography (HPLC) [7][8][9][10], the last one is the most used technique by pharmaceutical laboratories.…”
