Activation of Transcription by Recombinant Upstream Stimulatory Factor 1 Is Mediated by a Novel Positive Cofactor

Abstract: The transcription factor USF1 belongs to the family of basic helix-loop-helix proteins that are involved in the regulation of various important cellular processes. Here we characterized the factors involved in the activation of transcription by upstream stimulatory factor 1 (USF1) in a reconstituted class II gene transcription system. Activation of transcription by both wild type USF1 and a GAL-USF (amino acids 1-94 of the yeast activator protein GAL4 fused to amino acids 17-196 of USF) fusion protein required… Show more

“…Thus, it is possible that PSF could likewise increase binding of USF to element AB via a mechanism similar to that by which NonO enhances E47 binding. Similarly, the heatstable protein PC5 has been described as a cofactor that stimulates transcriptional activity of USF (43).…”

“…Human USF was first designated as major late transcription factor (16). Three USF isoforms have been described, USF1, 2a, and 2b, with apparent molecular masses of 43,44, and 38 kDa, respectively (23)(24)(25). USF1 and USF2a and 2b are encoded by two different genes, and USF2a and 2b are generated by differential splicing.…”

“…Preparation and purification of recombinant or native general transcription factors, TFIIA, TFIIB, TFIID, TFIIE, TFIIF, TFIIH, and RNA polymerase II, and the conditions used for the transcription reactions have been described previously (26,47,70). Transcriptions were conducted in the presence of 100 M all four nucleotides, and transcripts were analyzed by primer extension with an end-labeled primer complementary to the luciferase gene 28 bp downstream of the promoter cloning site with sequence CTTTATGTTTTTGGCGTCTTCCA essentially as described previously (32).…”

“…The ubiquitously expressed factor USF commonly consists of two subunits, USF-1 and USF-2 (26), and has been shown to be a transcriptional activator in combination with other factors (27)(28)(29)(30). For example, in the immunoglobulin heavy chain enhancer, activity is detected only in the presence of a three protein-DNA complex consisting of USF, PU.1, and Ets-1 (15).…”
