A Conserved Tissue-Specific Structure at a Human T-Cell Receptor β-Chain Core Promoter

Abstract: The T-cell receptor (TCR) ␤-chain promoters have been characterized as nonstructured basal promoters that carry a single conserved ubiquitous cyclic AMP-responsive element. Our investigation of the human TCR ␤ gene uncovers a surprisingly complex and tissue-specific structure at the TCR V␤ 8.1 promoter. The core of the promoter (positions ؊42 to ؉11) is recognized by the lymphoid cell-specific transcription factors Ets-1, LEF1, and AML1 as well as by CREB/ATF-1, as is demonstrated in gel shift and footprinting… Show more

“…The Ets factors that bind to DNA as monomers are rather weak activators of transcription and protein ± protein interaction with other factors has been shown to strengthen their transactivating potential, although in some cases it can also lead to transcriptional repression (Sharrocks et al, 1997). Cooperation between Ets and other factors facilitates binding to sites not matching preferred consensous binding site (Halle et al, 1997). For example, the GABP complex consists of an Ets family member, GABPa, and an ankyrin repeat-containing protein, GABPb.…”

“…Firefly luciferase activities were normalized to Renilla luciferase activities except in the case of CMV-Sp1, which significantly stimulated the TK promoter, which was co-transfected. In the latter case, the firefly luciferase activities were normalized to the protein content of each sample as described previously (25,26). The results show the mean values of three experiments with standard errors.…”

“…Although the V ␤ promoter is required for lineage-specific TCR␤ transcription, its role in regulating V ␤ gene rearrangement remains unclear (13)(14)(15)(16)(17). In addition to V ␤ promoters and E ␤ , there are likely other cis-regulatory elements involved in the control of various aspects of TCR␤ gene rearrangement and transcription.…”
