Activation of Protein Synthesis by Microsomes from Aging Beet Disks

Abstract: Suimmary. Microsomal fractions isolated from sterile, aged disks of red beetrool Jncorporate leucine into protein when supplemented with the supernatant fraction, ATP, GTP, and KCl; the incorporation is sensitive to RNase and is not duLe to 'bacteria. Thle nicrosomal activity is inhibited by puromycin and cycloheximide but is virtually insensitive to both D-threo and L-threo-chloramphenicol, as predicted from physiological studies.Microsomes isolated from fresh disks have much lower incorporatin;g ability than… Show more

“…The development of ion absorption capacity along with increased respiration has been suggested to involve basic alterations in physiological processes of plant tissue (1,9,14,20,40) and especially in protein metabolism (4,12,20,27). Figure 9A shows results of an experiment on the effect of cycloheximide, a known inhibitor of protein synthesis (4), on the development of K absorption capacity.…”

“…Bacteria have been shown to influence certain aspects of the aging of plant tissue (4,16); however, this influence appears to be negligible in bean stem slices (30). Nevertheless, 50 og ml-' chloramphenicol, an effective bacteriostat (16,40), was used routinely as a precautionary measure to maintain a low bacterial count in the aging solution.…”

“…Although differing in detail, this phenomenon has been described in a number of systems including seed embryos induced to germinate (13,15), fertilized sea urchin eggs (8), plant storage tissues activated by excision (5,10), and mammalian cell cultures in which growth is resumed by dilution (7,11,20,25). The details of the process seem to vary for the different situations.…”
