Antimicrobial Properties of Actinomycetes Isolated from Menengai Crater in Kenya

A study was carried out to isolate and screen actinomycetes for antimicrobials from Menengai Crater in Kenya. The actinomycetes were isolated using starch casein agar, Luria Bertani agar and starch nitrate agar. Primary screening for antagonism was carried out using perpendicular method while secondary screening was done using agar disk technique. Extraction of the antimicrobials was carried out using ethyl acetate. Sensitivity testing of the crude extracts against Staphylococcus aureus, Bacillus subtilis, Escherichia faecalis, Escherichia coli, Klebsiella pneumoniae, Salmonella typhi, Xanthomonas campestris, Erwinia carotovora, Candida albicans, Alternaria alternate and Fusarium oxysporum was carried out using agar well technique. Biochemical tests and carbon source requirements were used in characterization of the selected antimicrobial producers. M1 was the best agar medium for isolation of actinomycetes. The number of actinomycetes from regions A, B, C, and D in the crater varied significantly (F = 27.50 P = 0.000). Out of the 156 actinomycetes isolates, 20 isolates were positive for both primary and secondary screening for antimicrobials. There was no significant difference in the zones of inhibition in primary screening of the actinomycetes for antagonistic properties against the test pathogens (F = 1.6957 P = 0.0838). The zones of inhibition after secondary screening varied significantly (F = 2.4473 P = 0.0089). Likewise, there was a significant difference (F = 6.6046 P = 0.001338) in the zones of inhibition after exposing the pathogens to ethyl extracts of the selected antagonistic actinomycetes. There is need to purify and characterize the antimicrobials obtained from the present study.

Multi-drug resistant pathogens are a leading cause of human morbidity and mortality all over the world. This study aimed at isolating actinomycetes from the soils of Menengai crater and characterizing selected actinomycetes using morphological, biochemical, and molecular techniques. The study involved isolation of actinomycetes using Starch Casein (SC), Luria Bertani (M1) and Starch Nitrate (SN) agar media. The isolated actinomycetes were characterized using morphological and biochemical techniques. The screening for antibiotic activity was carried out via primary screening using perpendicular method and secondary screening using agar plug technique against Staphylococcus aureus (ATCC 25923), Streptococcus pneumoniae (ATCC 49617), Enterococcus faecalis (ATCC 29212), Escherichia coli (ATCC 25922), Pseudomonas aeruginosa (ATCC 27853), Proteus vulgaris (ATCC 49990), Alternaria citri (ATCC 1015), Candida albicans (ATCC 10231), Fusarium oxysporum (ATCC 16608) and Ustilago maydis (ATCC 14826). The best actinomycetes based on size of inhibition and broad spectrum of activity coded PAN 25, PAN 41, PAN 75 and PAN 110 were selected for further bioassay. Extraction of antibiotics was carried out using ethyl acetate. The selected actinomycetes were characterized using molecular technique that involved extraction of 16S rRNA gene, PCR and phylogenetic studies. The data obtained was analyzed using Statistical Package of Social Sciences (SPSS) version 25 software. The number of the isolated actinomycetes varied significantly (F=3.315 P=0.04218). There was no significant difference in the zones of inhibition between the isolates PAN 25, PAN 41, PAN 75 and PAN 110 (F=0.8928 P=0.5431). In addition, there was no significant difference in the zones of inhibition between the extracted antibiotics (F=1.726424 P=0.126281). The 16S rRNA gene sequence and nucleotide BLAST from NCBI revealed that isolate PAN 25 had similarity with Streptomyces variabilis, isolate PAN 41 (S. indiaensis), isolate PAN 75 (S. luteogriseus strain ZG728) and isolate PAN 110 (S. acrimycini strain K30). Menengai crater had actinomycetes that inhibited growth of the selected actinomycetes. There is need of isolating actinomycetes from other unique regions in Kenya. Large scale production of antibiotics from the selected actinomycetes need also to be carried out.

A novel halophilic actinobacterium strain, designated H8(T), was isolated from a Saharan soil sample collected in El-Goléa, South Algeria. Strain H8(T) was identified as representing a new genus using a polyphasic taxonomic approach. Phylogenetic analysis revealed that strain H8(T) shared the highest degree of 16S rRNA gene sequence similarity with 'Mzabimyces algeriensis' DSM 46680(T) (93.0 %), Saccharopolyspora ghardaiensis DSM 45606(T) (91.2 %), Halopolyspora alba DSM 45976(T) (90.8 %) and Actinopolyspora mortivallis DSM 44261(T) (90.0 %). The strain was found to grow optimally at 28-35 °C, at pH 6.0-7.0, and in the presence of 15-25 % (w/v) NaCl. The substrate mycelium was observed to be well developed and fragmented in liquid medium and on solid medium. The aerial mycelium was observed to be moderately abundant and to form long chains with non-motile, smooth-surfaced and ovoid or spherical spores at maturity. The cell wall of strain H8(T) was found to contain meso-diaminopimelic acid. The whole-cell hydrolysates were found to mainly contain arabinose and galactose. The diagnostic phospholipid detected was phosphatidylcholine, and MK-9(H4), MK-9(H2) and MK-10(H2) were found to be the predominant menaquinones. The major cellular fatty acids were determined to be anteiso-C17:0 and iso-C15:0. The genomic DNA G+C content of strain H8(T) was determined to be 71.3 mol%. The genotypic and phenotypic data showed that the strain represents a novel genus and species, for which the name Bounagaea algeriensis gen. nov., sp. nov. is proposed, with the type strain H8(T) (=DSM 45966(T) = CECT 8470(T)).
