Acid cleavable surface enhanced raman tagging for protein detection

Abstract: Dye conjugation is a common strategy improving the surface enhanced Raman detection sensitivity of biomolecules. Reported is a proof-of-concept study of a novel surface enhanced Raman spectroscopic tagging strategy termed as acid-cleavable SERS tag (ACST) method. Using Rhodamine B as the starting material, we prepared the first ACST prototype that consisted of, from the distal end, a SERS tag moiety (STM), an acid-cleavable linker, and a protein reactive moiety. Complete acid cleavage of the ACST tags was achi… Show more

“…detection and identication of nucleic acids, lipids, peptides, proteins, etc. [36][37][38] Herein, we have investigated the metal-molecule interaction of one such organoselenium compound, selenourea (SeU) adsorbed over Ag and Au NPs and also in nanoassemblies where the molecule is sandwiched between Ag and Au NPs. SeU belongs to a class of diamides that are primarily used in the synthesis of selenium heterocycles that exhibit anti-inammatory and anti-tumor activities, among other medicinal uses.…”

“…In SERS, the enhancement of Raman intensity occurs mainly due to the electromagnetic and the chemical effect . Recently, SERS spectroscopy has been widely used as a promising tool for the detection/sensing of various organic and biological molecules with higher sensitivity and precision . This technique has also been exploited to study surface‐induced catalytic reactions .…”
