Acid–base titration using a microfluidic thread-based analytical device (μTAD)

Abstract: This work presents a novel analytical approach for precise and quick (within 2 minutes) determination of acid and base concentrations (of very small amount) by titration using a microfluidic thread-based analytical device (μTAD).

“…Moreover, distance and barcode-based detection are another two possible low-cost detection strategies (Tan et al, 2021). Distance-based detection relies on the fact that disparate wetting performances can be induced by different analytes for identification (Alsaeed and Mansour, 2020;Jarujamrus et al, 2020;Shimazu et al, 2022). Moreover, barcode detection can provide results of multiple analyte reactions (e.g., blood typing) that otherwise are difficult to achieve (Nilghaz et al, 2014).…”

“…The base processes of operation in the μTAD are driven by capillary action between the threads that drive the sample to the sites where reagents are immobilized so as to promote a reaction . These reagents can be designed to leverage detection techniques such as colorimetry, which provides a qualitative signal to the eye or to a smartphone-based detection, which are further highlighted by the thread's white color. Owing to their utility, μTADs have been used to perform a myriad of assays including glucose assays, enzyme-linked immune-sorbent assays (ELISA), immunochromatographic assays. …”

“…However, pH based titrimetric analyses indicate noting the titrimetric value of a titration at the turning point of the color change of the indicators. [51][52][53] Hence, based on this principle of pH-dependent color change, we have integrated thread substrate with a colorimetric microbial assay by employing the conventional metabolic sugar fermenting prole of microbes [54][55][56] (Fig. 1a).…”
