Acetolactate synthase activity and chlorsulfuron sensitivity of gamma-irradiated lentil (Lens culinaris Medik.) cultivars

Abstract: SU MMARYLentil (Lens culinaris Medik.) cultivars (Jordan 1 and Jordan 2) were sensitized to chlorsulfuron herbicide. Both cultivars were subjected to three doses of gamma-irradiation (90, 100 and 110 Gray) to develop plants tolerant/resistant to this herbicide. The herbicide-tolerant plants as well as the sensitized plants were subjected to acetolactate synthase (ALS) enzyme assay to reveal the biochemical basis of plant tolerance. The results indicated that as the radiation dose increased, the tolerance to th… Show more

“…These mutagens can be effectively used to generate mutations in lentil that ultimately produces resistance in plants against herbicides. Many other studies reported that induced mutations are efficient in creation of herbicide resistant mutants in different crops [ 14 , 16 , 25 , 26 ].…”

“…Chant (2004) [ 26 ] developed an imidazolinone tolerant lentil mutant (RH44) by EMS mutagenesis. Malkawi et al ., (2003) [ 25 ] treated lentil cultivars with gamma radiations to develop tolerance against chlorsulfuron herbicide. Similarly, Sebastian and Chaleff (1987) [ 6 ] selected four mutants of soybean (1-126A, 1-166A, 1-183A and 1-184A) which appeared to be sufficiently tolerant to chlorsulfuron herbicide.…”

“…After centrifugation, the GSH content in the supernatant of the maize root was measured spectrophotometrically at 412 nm in the presence of DTNB and calculated via a comparison with a known GSH concentration. 42 2.7 Determination of ALS activity ALS activity was determined according to the method of Malkawi et al 43 Acetolactate was measured by spectrophotometry (525 nm) and calculated by comparison against known acetolactate concentrations. Protein content was determined according to the Bradford method using bovine serum albumin as the reference material.…”

“…To investigate the effect of safener to target enzyme, ALS activity was determined as described previously (Malkawi et al, 2003). ALS activity was expressed as amount of acetoin composed from acetolactate catalyzed by ALS per hour per milligram of enzyme (nmol h -1 mg -1 protein).…”
