Accession-specific modifiers act with ZWILLE/ARGONAUTE10 to maintain shoot meristem stem cells during embryogenesis in Arabidopsis

Abstract: BackgroundStem cells located in the centre of the shoot apical meristem are required for the repetitive formation of new organs such as leaves, branches and flowers. In Arabidopsis thaliana, the ZWILLE/PINHEAD/AGO10 (ZLL) gene encodes a member of the ARGONAUTE (AGO) protein family and is required to maintain shoot meristem stem cells during embryogenesis. In the Landsberg erecta (Ler) acession, ZLL is essential for stem cell maintenance, whereas in the Columbia (Col) accession its requirement appears masked by… Show more

“…Very recently, a RNA-based sequence analysis revealed an upregulation (over eightfolds) of germin-like protein genes (ZmGLP2-1) during maize embryogenic tissue culture initiation (Salvo et al 2014). In Arabidopsis, the germin-like protein 6 (AT5G39100) and a putative GLP (AT5G38960) were differentially expressed in torpedo stage embryos (Tucker et al 2013). Arabidopsis calmodulin N-methyltransferase (CaM KMT), involved in transfer of methyl groups to calmodulin (CaM), was highly expressed in the apical meristemic region of plants during active growth stages (Banerjee et al 2013).…”

“…In particular, the mutation in ZWILLE / PINHEAD / AGO10 ( ZLL ) shows defects in maintenance of embryonic shoot apical meristem. However, these defects were apparent in Ler, but not in Col-0, because the defects were masked by the modifiers in Col-0 ( Tucker et al, 2013 ). One of the modifiers in the ZLL function appeared to be the Arabidopsis Cyclophilin-40 orthologue SQUINT (SQN, AT2G15790) ( Tucker et al, 2013 ).…”

One of the modifiers in the ZLL function appeared to be the Arabidopsis Cyclophilin-40 orthologue SQUINT (SQN, AT2G15790) ( Tucker et al, 2013 ). In addition, the altered meristem program ( amp1 ) mutation differently affected the flowering time in Col-0 and Ler ( Lee, 2009 ), presumably because of the different genetic modifiers in these accessions.…”

“…In the L er accession that has higher SQN expression levels and thus a higher potential for AGO1-mediated slicing of HD-ZIP III mRNAs, the decoy function of ZLL/AGO10 is crucial for shoot meristem stem cell maintenance during embryogenesis. By contrast, in the Columbia accession that has lower SQN expression, ZLL/AGO10 is dispensable for this process ( Tucker et al., 2013 ).…”
