Abstract A31: Activation of the RAS-MAPK pathway in primary neuroblastoma tumors is associated with poor prognosis

Abstract: We recently identified that mutations affecting the RAS-MAPK pathway occur frequently in relapsed neuroblastoma, which suggests that activation of this pathway may play an important role in the aggressive phenotype of these tumors. Furthermore, it was shown that the presence of these mutations may serve as a biomarker for treatment with MEK inhibitors both in vitro and in vivo. These results suggest that MEK inhibitors may be a viable treatment option for aggressive neuroblastoma tumors and warrant further inv… Show more

“…The MAPK signaling pathway also participates in the cellular activity of survival or death ( 28 ). Activation of the RAS-MAPK pathway is associated with poor prognosis in neuroblastoma tumors ( 29 ). In the present study, it was therefore inferred that PRKCG may affect the prognosis of GBM by the influencing MAPK signaling pathway.…”

“…To further shortlist which of these 13 genes could have the highest potential as a target for combinatorial immunotherapeutic approaches, we validated the correlation between their gene expression and the immune cell cytotoxicity score in two additional large, independent bulk-RNA sequencing datasets of 498 (SEQC) and 122 (COMBAT-Versteeg) neuroblastoma tumors 47,48 . Out of the 13 genes, Macrophage Migration Inhibitory Factor (MIF) and Midkine (MDK) negatively correlated with cytotoxicity in both datasets (Fig.…”
