Absorption changes of Photosystem II donors and acceptors in algal cells

Abstract: Flash‐induced absorption changes were measured in the 390–450 nm region, using whole cells of a double mutant strain of Chlorella sorokiniana, lacking PS I centers and part of the pigment antenna. Spectral changes associated with PS II acceptors (Qa and Qb) and donors (p680, Z and L) are described. In this region, the spectrum obtained for Qb −/Qb differs markedly from that of Qa −/Qa. Different spectra were also obtained for the secondary donor (Z+/Z) when measured after one or three flashes.

“…The largest extent at 443 nm, attributable to electrochromism, was observed with oxygen-evolving material (Figure 2, blue, solid line). Its difference spectrum was similar to previously reported ones (4,40,47,48,59,60). With Mn-depleted centers at pH 9 the spectral shape was similar to the former ones; however, the extent was about halved (Figure 2, blue, dotted line).…”

“…This interpretation was confirmed by Saygin and Witt (1985) but challenged by Lavergne (1984Lavergne ( , 1986. The observed oscillation pattern of absorbance changes at 920 nm with flash number was compatible with assignment to the S2-state (Dismukes and Mathis, 1984).…”

“…Addition of p-benzoquinone, a protein cross-linker, blocks overall cell metabolism without stopping the primary photosynthetic reactions at the level of the thylakoid membrane. It results in a full oxidation of the plastoquinone pool in the dark (24,25), which we show here to considerably slow the reducing flux (slower than 0.1 s Ϫ1 , see Fig. 2A).…”
