The liver is the most central organ and the largest gland of the body that influences and controls a variety of metabolic and catabolic processes. It produces inconceivable many essential proteins, is responsible for the recovery of various food components, degrades toxins, mediates the bile production, and is involved in the excretion of unwanted metabolites. Several of these anabolic or catabolic functions of the liver depend on trace elements. These are either integral part of enzymes, cofactors, or act as chemical catalysts. Therefore, a lack of trace elements can lead to organ failure or systemic illness. Conversely, excessive hepatic trace element deposition resulting from genetic disorders, intoxication, extensive dietary supply, or long-term parenteral nutrition may cause hepatic inflammation, fibrosis, cirrhosis, and even hepatocellular carcinoma. Although specific serum parameters currently allow rough assessment of metal deficit and excess, the precise quantification of hepatic metal content in liver is presently only possible by different titration or staining techniques of biopsy specimens. Recently, novel innovative metal imaging techniques were developed that are on the way to replace these traditional methods. In the present review, we summarize the function of different trace elements in liver health and disease and discuss the present knowledge on how quantitative biometal imaging techniques such as synchrotron X-ray fluorescence microscopy, secondary ion mass spectrometry, and laser ablation inductively coupled plasma mass spectrometry enrich diagnostics in the detection and quantification of hepatic metal disorders. We will further discuss sample preparation, sensitivity, spatial resolution, specificity, quantification strategies, and potential future applications of metal bioimaging in experimental research and clinical daily routine. © 2015 Wiley Periodicals, Inc. LA-ICP-MS allows precise, relatively fast, and spatially resolved measurements of elements and isotope ratios at trace and ultratrace concentration levels with minimal sample preparation. Over the past few years this technique has undergone rapid development, and it has been increasingly applied in many different fields, including biological and medical research. The analysis of essential, toxic, and therapeutic metals, metalloids, and nonmetals in biomedical tissues is a key task in the life sciences today, and LA-ICP-MS has proven to be an excellent complement to the organic MS techniques that are much more commonly employed in the biomedical field. In order to provide an appraisal of the fast progress that is occurring in this field, this review critically describes new developments for LA-ICP-MS as well as the most important applications of LA-ICP-MS, with particular emphasis placed on the quantitative imaging of elements in biological tissues, the analysis of heteroatom-tagged proteins after their separation and purification by gel electrophoresis, and the analysis of proteins that do not naturally have ICP-MS-detectable elements in their structures, thus necessitating the use of labelling strategies.

Abstract. About 30 % of all proteins contain metals either as cofactor or covalently bound as part of the protein.Some of these metalloproteins such as transferrin (TRF) and haemoglobin (HGB) are regularly measured in clinical laboratories. However, for most of them, no primary reference measurement procedures exist, although directives such as the EC-directive covering in vitro diagnostic medical devices ((IVD-) directive 98/79/EC) and standards such as EN ISO 17511:2003 demand the traceability of results received for analytes in samples of human origin. The EMRP project HLT05 aims at establishing a traceability chain for metalloproteins as is already in place for some inorganic and small organic molecules in biological samples.Environ 30 % de toutes les protéines contiennent des métaux, soit en tant que cofacteur, soit faisant partie de la protéine par liaison covalente. Certaines de ces métalloprotéines, telles que la transferrine (TRF) et l'hémoglobine (HGB), sont régulièrement analysées en laboratoires cliniques. Cependant, pour la plupart d'entre elles, il n'existe pas de méthode de mesure de référence primaire bien que des directives, telles que la directive CE relative aux dispositifs médicaux de diagnostic in vitro (directive (IVD) 98/79/CE), et des normes, telles que l'EN ISO 17511:2003, exigent la traçabilité des résultats pour les analytes dans les échantillons d'origine humaine. Le projet HLT05 de l'EMRP a pour objectif de mettre en place pour les métalloprotéines une chaîne de traçabilité similaire à celle existant déjà pour les molécules inorganiques et les petites molécules organiques dans les échantillons biologiques.
