Abscisic Acid Inhibition of Endosperm Cell Division in Cultured Maize Kernels

Abstract: The response of developing maize (Zea mays L.) endosperm to elevated levels of abscisic acid (ABA) was investigated. Maize kernels and subtending cob sections were excised at 5 days after pollination (DAP) and placed in culture with or without 90 micromolar (±)-ABA in the medium. A decreased number of cells per endosperm was observed at 10 DAP (and later sampling times) in kernels cultured in medium containing ABA from 5 DAP, and in kernels transferred at 8 DAP to medium containing ABA, but not in kernels tran… Show more

“…There are several, possibly interacting, factors contributing to the variation in grain weight. It may result from intra-panicle and intra-spikelet differences in development rate and time of pollination that are reflected in duration of grain-filling period (Bonnett 1961a, Tibelius andKlinck 1987), vascular transport capacity (Hanif and Langer 1972, shown in wheat;Housley and Peterson 1982), cell division rate and duration of developing grains as shown for wheat (Brocklehurst 1977, Radley 1978, and/or hormonal balance as shown for barley and maize (Michael and Seiler-Kelbitch 1972, Mounler et al 1980, Myers et al 1990, Ober et al 1991.…”

“…Endosperms were dissected out and treated with pectinase to obtain a homogeneous suspension of nuclei as previously described (13). Nuclei were stained with acetocarmine and counted on a hemacytometer (X400; Improved Neubauer Ultra Plane, Model 3500, Hausser Scientific, Blue Bell, PA).…”

“…Explants of whole kernels and subtending cob sections were cultured at 5 DAP as previously described (13). Combinations in an array of three sucrose concentrations (0.29, 0.44, or 0.58 M) and three sorbitol concentrations (0, 0.15 or 0.29 M) were used to obtain six media at three levels of Ow (Table I).…”
