A Validated RP HPLC Method for Estimation of Almotriptan Malate in Pharmaceutical Dosage Form

Abstract: An accurate, sensitive, precise and robust reverse phase high performance liquid chromatographic method for the estimation of almotriptan malate in pharmaceutical dosage form has been developed and validated. Chromatographic separation was conducted on Phenomenex Gemini C18 (250 mm´4.6 mm, 5m) column at room temperature using potassium dihydrogen phosphate buffer: acetonitrile (80:20) as a mobile phase, pH adjusted to 3.2 ± 0.1 with 10% sodium hydroxide and at a flow rate of 1.5 mL min -1 , while UV detection … Show more

“…A few HPLC and HPTLC methods 15–21 as well as spectrophotometric methods 22–24 are reported for the determination of ALM in pharmaceutical formulations or biological fluids. The reported chromatographic methods are expensive and time‐consuming while the spectrophotometric methods are either less sensitive or involve heating.…”

“…Forner et al [2] and Bosch et al [3] have synthesised almotriptan malate and have published the data in US5565447. Literature review suggested that, several analytical methods such as HPLC [4][5][6][7], HPTLC [8], HPLC-MS/MS [9], LC-ESI-MS/MS [10] are available for the estimation of drug content. Analytical method validation ensures that various HPLC analytical techniques shall give reliable and repeatable results; it is a crucial step in developing new dosage forms as it provides information about accuracy, linearity, precision, detection, and quantitation limits.…”

“…Almotriptan is given orally as the malate, and doses are expressed in terms of the base; almotriptan malate 8.75 mg is equivalent to about 6.25 mg of almotriptan. 1 Few analytical methods have been reported for its determination including spectrophotometry, 2-6 spectrouorimetry, 3 HPLC [7][8][9] and LC-MS. 10 Zolmitriptan (ZOL), chemically known as (S)-4-{3-[2-(dimethylamino)ethyl]indol-5-ylmethyl}-1,3-oxazolidin-2-one (Fig. 1), is also a selective serotonin (5-HT1) agonist with actions and uses similar to those of ALM.…”
