A Telomerase‐Responsive DNA Icosahedron for Precise Delivery of Platinum Nanodrugs to Cisplatin‐Resistant Cancer

Abstract: A telomerase-responsive DNA icosahedron was designed to precisely release caged platinum nanodrugs into cisplatin-resistance tumor cells for effective therapy. This DNA icosahedron was constructed from two pyramidal DNA cages connected with telomerase primers and telomeric repeats, and platinum nanodrugs were then encapsulated into the DNA structure. In the presence of telomerase, the primers are extended, leading to inner-chain substitution of the DNA icosahedron and subsequent release of the caged nanodrugs.… Show more

“…Logic‐gated designs may be integrated into the locking mechanism to enable cargo releasing in the presence of multiple cellular targets at the same time (and gate), minimizing false‐positive events. Gu et al constructed a telomerase‐responsive DNA icosahedral structure loaded with platinum nanodrugs (Figure c) . Telomerase is reportedly overexpressed in the majority of cancer cells while nearly absent in normal cells.…”

“…In another example, connective regions of DNA icosahedra are designed to include telomerase primer and telomeric repeats. Telomerase activity on these carriers results in primer elongation and destabilization of sticky ends, causing dissociation of the DNA icosahedron and release of the encapsulated platinum nanoparticles …”
