A statherin-derived peptide promotes hydroxyapatite crystallization and in situ remineralization of artificial enamel caries

Abstract: A novel biomimetic peptide inspired by salivary statherin was developed and exhibited beneficial potentials in promoting remineralization of hydroxyapatite on tooth enamel surface, providing a desirable alternative restorative strategy against dental caries.

“…A number of synthetic peptides, especially those derived from proteins in an enamel or dentin matrix, have been developed to promote remineralization of enamel and dentin [ 34 , 35 , 36 , 37 ]. Most were capable of inducing hydroxyapatite deposition, and a DSP-derived peptide was reported to act on pulp cells and induce osteodentin formation [ 38 ].…”

“…Human oral keratinocytes (HOKs) (ATCC11303) were chosen to assess cell viability, which were derived from the stratum corneum of the oral mucosa, in order to study the cytotoxic effect of TDP (Liu et al, 2018; Wang et al, 2018). HOKs were cultured in alpha Dulbecco's Modified Eagle Medium containing 10% fetal bovine serum and 1% penicillin–streptomycin.…”

“…The concentrations of the peptide, TVH-19, in the solution, before and after incubation, were evaluated using a micro bicinchoninic acid (BCA) Protein Assay Kit (CWBio, Beijing, China) to calculate the amount of peptide bound to the unit surface area of the HA powder. Linear adsorption isotherms were then generated according to the Langmuir Equation (1) [32,33]. The maximum number of adsorption sites available for the protein per unit of the HA surface area and the affinity of the protein molecules for the HA adsorption sites were obtained from the resulting best-fit line:…”
