A Starburst‐Shaped Heterometallic Compound Incorporating Six Densely Packed Gd3+ Ions

Abstract: The heterotritopic ligand [bpy(DTTA)2]8- has two diethylenediamine-tetraacetate units for selective lanthanide(III) coordination and one bipyridine function for selective Fe(II) coordination. In aqueous solution and in the presence of these metals, the ligand is capable of self-assembly to form a rigid supramolecular metallostar structure, [Fe[Gd2bpy(DTTA)2(H2O)4]3]4-. We report here the physicochemical characterization of the dinuclear complex [Gd2bpy(DTTA)2(H2O)4]2- and the metallostar [Fe[Gd2bpy(DTTA)2(H2O)… Show more

“…The attachment of current commercially available contrast agents such as [Gd (DOTA)] 2-and [Gd(DTPA)] 2-to macromolecular constructs has been extensively studied, and in several cases, enhancements have been observed upon slowing of molecular rotation. [71][72][73][74][75][76][77][78][79] These contrast agents, however, are somewhat restricted because they do not have the optimal water-exchange rates that would lead to large enhancements in relaxivity. The major advantage of these aminocarboxylate-based contrast agents is their high watersolubility.…”

“…The former shows an increase of about 200% at 40 MHz on the formation of the heterobimetallic complex (metallostar), giving rise to an exceptionally high relaxivity confined to a small molecular space. 39,40 This high in vitro relaxivity is retained under in vivo conditions in mice. The pharmacokinetics of this metallostar, injected intravenously, were found to be similar to that of Gd-DOTA, involving fast renal clearance and a leakage to the extracellular space in the muscle tissue.…”
