Plant‐parasitic nematode species are one of the greatest threats to cause serious yield loss throughout soybean production regions of the United States. Soybean [Glycine max (L.) Merr.] cultivars of different maturity groups carry known resistance to soybean cyst nematode (Heterodera glycines, SCN) at rgh1 and Rhg4 loci. Continuous use of these genes caused SCN population shifts and overcoming host resistance over time. Lack of genetic diversity created a bottleneck in development of new cultivars containing novel SCN resistance genes. Additional sources of resistance to southern root‐knot nematode (Meloidogyne incognita, SRKN) and reniform nematode (Rotylenchlus reniformis, RN) could result in supplemental protection of soybean yield. This research focused on determining known and new sources of SCN resistance in 1220 soybean accessions divided into two groups: (i) early‐maturity groups (000‐II) screened with two SCN populations, and (ii) medium‐maturity groups (III‐V) screened with six SCN populations. Based on the initial screening results, a subset of 76 accessions was selected for further phenotyping with SCN, SRKN, and RN populations and supplemented by genotyping using molecular markers. The results revealed that 56 lines were resistant to two nematode species in various combinations, and 12 lines were resistant to all three nematode species. Moreover, potential new sources of resistance‐carrying genes other than rhg1 and Rhg4 loci were pinpointed. Interestingly, RN resistance highly correlated with rhg1 locus suggesting a common gene or tight linkage. Identified resistant soybean accessions provide promising materials for discovery of novel genes to support cultivar development.

Genes functioning in membrane fusion were originally identified genetically in Saccharomyces cerevisiae and are found in all eukaryotes. Components of the unit, soluble N-ethylmaleimidesensitive fusion protein attachment protein receptor (SNARE), function in the plant genetic model Arabidopsis thaliana during its defense to shoot pathogens. Regarding defense, little is understood about SNARE in roots or its regulation. Experiments in Glycine max (soybean) have provided an opportunity to perform such studies, revealing that SNARE genes are expressed under natural conditions in root cells undergoing defense to parasitism by the nematode Heterodera glycines. Presented here, the G. max homolog of S. cerevisiae suppressor of sec1 (SSO1), identified genetically in A. thaliana as PENETRATION1 (PEN1) and named in its genomic annotation as syntaxin 121 (SYP121) functions in the resistance of G. max to H. glycines. Genetic experiments demonstrate Gm-SYP121 is co-expressed with homologs of other SNARE genes exhibiting measurable transcript levels in infected cells undergoing resistance. These genes include synaptosomal-associated protein 25, homologous to A. thaliana SNAP33 (SNAP-25/SNAP33/SEC9); mammalian uncoordinated-18 (MUNC18/SEC1); synaptotagmin/tricalbin-3 (SYT/TCB3); synaptobrevin/vesicle associated membrane protein/YKT6/SEC22 (SYB/VAMP/YKT6/SEC22); N-ethylmaleimide-sensitive fusion protein (NSF/SEC18) and alpha-soluble N-ethylmaleimide-sensitive fusion protein associated protein (α-SNAP/SEC17). Experiments show each SNARE component functions in resistance. In contrast, a coatomer zeta/ retrieval3 (Cζ/RET3) homolog known to function in retrograde transport within and between the Golgi and endoplasmic reticulum does not appear to function in resistance. Experiments show that SNARE is co-regulated along with a β-glucosidase having homology to PEN2 and an ATP binding cassette transporter exhibiting homology to PEN3. ARTICLE HISTORY

Nevertheless, the genetic networks underlying gene expression control in the syncytium are poorly understood. DNA methylation is a critical epigenetic mark that plays a key role in regulating gene expression. To determine the extent to which DNA methylation is altered in soybean (Glycine max) roots during the susceptible interaction with SCN, we generated whole-genome cytosine methylation maps at single-nucleotide resolution. The methylome analysis revealed that SCN induces hypomethylation to a much higher extent than hypermethylation. We identified 2,465 differentially hypermethylated regions and 4,692 hypomethylated regions in the infected roots compared with the noninfected control. In addition, 703 and 1,346 unique genes were identified as overlapping with hyper-or hypomethylated regions, respectively. The differential methylation in genes apparently occurs independently of gene size and GC content but exhibits strong preference for recently duplicated paralogs. Furthermore, a set of 278 genes was identified as specifically syncytium differentially methylated genes. Of these, we found genes associated with epigenetic regulation, phytohormone signaling, cell wall architecture, signal transduction, and ubiquitination. This study provides, to our knowledge, new evidence that differential methylation is part of the regulatory mechanisms controlling gene expression changes in the nematode-induced syncytium.
