A simplified and accurate method for the analysis of urinary metabolites of testosterone‐related steroids using gas chromatography/combustion/isotope ratio mass spectrometry

Abstract: These carefully controlled analytical conditions are compatible with routine operations, affording accurate and precise results for the more diagnostically relevant metabolites such as testosterone itself and the 5α- and 5β-androstanediols. The values of the TS-ERC pairs measured in reference populations are described and the results from the routine testing of several hundreds of athletes' samples are discussed. Robust, this technique permitted the detection of adverse findings that would have been missed had… Show more

“…The compounds monitored were all the compounds included in the steroid module of the ABP. The 13 C delta values of the target compounds (TC) testosterone (T), epitestosterone (E), androsterone (A), etiocholanolone (Etio), 5α-androstan-3α,17β-diol (5aAdiol), 5β-androstan-3α,17βdiol (5bAdiol) were determined. [12] Briefly, 3 to 6mL of urine depending on the concentrations established after the application of the method for the establishment of the SP were extracted as previously described.…”

“…Nowadays, a methodology based on determining the percentage of 13 C in the molecule, namely, GC with combustion isotope ratio mass spectrometry (GC-C-IRMS), is normally applied to ascertain if an atypical content of endogenous steroids is due to either its intake by an athlete or physiological factors [7,8].…”

“…The described confirmatory methods [12][13][14][15][16] are laborious, requiring a purification step by liquid chromatography in order to guarantee extracts of adequate purity suitable for IRMS analysis, precluding the application of the method on a large scale.…”
