A Simple, Sensitive, Rapid and Specific Detection Method for Bisphenol A based on Fluorescence Polarization Immunoassay

Abstract: A rapid, sensitive, specific and simple method based on Fluorescence Polarization Immunoassay (FPIA) was developed for the quantitative detection of Bisphenol A in water. 4, 4-Bis (4-hydroxyphenyl) valeric acid (BHPVA) was selected as the hapten to produce the immunogen. Fluorescein-labeled Bisphenol A derivatives (tracers) with different structures were synthesized and purified by thin layer chromatography. Based on the polyclonal antibody and tracers, an optimized FPIA method was developed with a detection l… Show more

“…Generally, high-performance liquid chromatography (HPLC) and gas chromatography (GC) are accurate and reliable but expensive, time-consuming and unsuitable for analysis of a large amount of sample. 22 Therefore, there is a big demand to develop another sensitive FPIA for determination of BPA based on a new uorescein and sensitive monoclonal antibody. ELISA 19 and CLEIA 1 are heterogeneous methods which need 2-3 h and several times of washing, while FPIA is a homogeneous method which needs no separation or washing steps but just a short time of 10 min.…”

“…20,21 Wu et al had set up a FPIA method based on uorescein (BVA-EDF) and polyclonal antibody, and the paper was published on Immunological Investigations. 22 Therefore, there is a big demand to develop another sensitive FPIA for determination of BPA based on a new uorescein and sensitive monoclonal antibody.…”

“…The anti-bisphenol A mouse monoclonal antibody was provided by the lab of Professor Chuanlai Xu [18] (School of Food Science & Technology, State Key Lab of Food Science and Technology, Jiangnan University, Wuxi, Jiangsu, China), dissolved in a PBS-based buffer (7 mg/mL in 0.01 M PBS, pH 7.4, 1% BSA, 1% glycerol, 0.02% azide). The polyclonal goat anti-mouse HRP-labeled antibody (clone A4416) (whole molecule, 0.5-3 mg/mL in 0.01 M PBS, pH 7.4, 1% BSA, 0.02% azide) and sheep anti-mouse IgG, 0.6-2 mg/mL in 0.01 M PBS, pH 7.4, 1% BSA, 0.02% azide) were provided by Merck Millipore (Darmstadt, Germany).…”

“…A suitable binding of antibody with antigen can be easily affected by chemicals that are structurally related to analytes, causing false positive results. 36,38 A set of important TBBPA analogues such as tetrabromobisphenol A-bis(2,3-dibromopropylether) (TBBPA-DBPE), 2,2 0 ,6,6 0 -tetrabromobisphenol A diallyl ether (TBBPA-BAE), hexabromocyclododecane (HBCD), 1,2-bis(pentabromodiphenyl)ethane (DBDPE), polybrominated diphenyl ethers (PBDEs) and bisphenol A (BPA) were therefore used to evaluate the cross-reactivity. The FPIA exhibited little cross-reactivity with some tested compounds including BDE-99 (3.8%), BDE-47 (4.2%), TBBPA-DBPE (6.0%), TBBPA-BAE (7.1%) and BPA (8.2%) (Table 2).…”
