A simple and efficient sequential electrokinetic and hydrodynamic injections in micellar electrokinetic chromatography method for quantification of anticancer drug 5‐fluorouracil and its metabolite in human plasma

Abstract: A simple and sensitive preconcentration strategy using sequential electrokinetic and hydrodynamic injection modes in micellar electrokinetic chromatography with diode array detection was developed and applied for the separation and determination of anticancer agent, 5‐fluorouracil and its metabolite, 5‐fluoro‐2′‐deoxyuridine, in human plasma. Sequential injection modes with increased analyte loading capacity using the anionic pseudo‐stationary phase facilitated collection of the dispersed neutral and charged a… Show more

“…In micellar electrokinetic chromatography (MEKC), analytes are separated by the differential partition between the pseudo-stationary phase and the surrounding electrolyte [21]. MEKC can also combine different stacking techniques to separate analytes simultaneously according to the properties of the analytes [22], and LVSS is one of the typical representatives [23].…”

“…However, the resolution decreased when SDS concentration was further increased to beyond 40 mM. This might be because the high concentration of SDS could result in strong interaction between micelle and analytes [ 28 ], which reduced the separation efficiency. In consideration of the resolution and peak shape, 40 mM was selected as the optimal concentration for SDS.…”
