A Shortcut to the Optimization of Cellulase Production Using the Mutant Trichoderma reesei YC-108

Abstract: Trichoderma reesei YC-108, a strain isolated by a kind of newly invented plate was found to over produce cellulase and it was then used as a cellulase producer. To get the maximum amount of cellulase, the combination of the medium ingredients, which has a profound influence on metabolic pathway was optimized using response surface methodology. The optimum composition was found to be 24.63 g/L wheat bran, 30.78 g/L avicel, and 19.16 g/L soya-bean cake powder. By using the optimized medium, the filter paper acti… Show more

“…To the best of our knowledge, this is the first report suggesting that MM + wheat bran-2 medium could increase the nematicidal activity of T6 culture filtrates against H. avenae J2s. This is consistent with the previous study of the fermentation medium and fermentation parameters in enhancing the bio-control ability of T. viride YC-108 mutant strain (Yan et al, 2012;Yang et al, 2016). Yan et al (2012) reported that wheat bran, avicel, and soya-bean cake powder, nitrogen source, and soybean meal could promote and increase the cellulase production from the strain of T. reesei YC-108.…”

“…The filter paper activity from mutants obtained with variation of cellulosic substrates ranging from 0.73 to 1.09 FPU/mL. Yan et al (2012) reported that cellulase production using the mutant T. reesei YC-108 increased the FPU activity nearly five times to 15.82 IU/mL. Meanwhile, the mutan NU-6 showed approximately 1.8 fold increase activity of FPase (4.7 U/mL) after the strain of T. reesei Rut C-30 was treatment with nitrosoguanidine followed by UV irradiation (Jun et al, 2009).…”

“…The quadratic model has high F value (21.01) and low P value (0.0064). Sathish et al [8] and Yan et al [14] also noticed that quadratic model is best suitable model for analysis of the mixture design. The special cubic and cubic models have the higher R [11] the model which have the higher R value and lower P value are significant based on this the cubic and special cubic models were rejected and further analysis was carried out with the quadratic model only.…”
