A Selective N-Type Ca2+-Channel Blocker Prevents CA1 Injury 24 h following Severe Forebrain Ischemia and Reduces Infarction following Focal Ischemia

Abstract: Summary: SNX-III (NEUREX Corporation, Menlo Park, CA, U.S.A.) an w-conopeptide, was tested for cy toprotection following normothermic ischemia using both a four-vessel occlusion model of severe forebrain isch emia and a model of transient middle cerebral artery oc clusion focal ischemia. Adult male Wi star rats were sub jected to 10 min of forebrain ischemia followed by 7 days of reperfusion. A single dose of SNX-Ill (5 mg/kg) was injected intravenously following delays of either 6 or 24 h after reperfusion. F… Show more

“…30,31 Similarly, the -conotoxin SNX-111, which potently blocks N-type Ca 2ϩ channels and thereby prevents glutamate release, has also been found to markedly reduce CA1 injury. 32,33 We explored whether the potent neuroprotective drugs NBQX and SNX-111 would provide enduring CA1 protection. Some of these data have previously been reported in abstract form.…”

“…Presynaptic N-type Ca 2 + channels playa crucial role in vesicular neurotransmitter release (KAMIYA et al1988;DUTAR et al1989), and drugs selective for this channel subtype attenuated neuronal injury in rodent cerebral ischemia studies (VALENTINO et al 1993;BUCHAN et al 1994). P-and Qtype channels, also expressed presynaptically, regulate physiological glutamate transmission in hippocampal slices (WHEELER et al 1994), and a peptide inhibitor of these channels reduced infarct volume following rodent cerebral ischemia (ASAKURA et al1997).…”
