A second proteinase encoded by a plant potyvirus genome.

Abstract: The RNA genome of tobacco etch virus (TEV) encodes a large polyprotein precursor that is processed to mature proteins by virus‐specific proteinases. Cleavage sites located within the carboxyl‐terminal two‐thirds of the polyprotein are processed by a TEV‐encoded 49 kd proteinase, while the enzyme(s) responsible for cleaving the remaining sites has not been found. In this study, a second TEV‐encoded proteinase has been identified based on cell‐free expression of defined RNA transcripts. The boundaries of this pr… Show more

“…1), similar to those characterized for tobacco etch virus (TEV [9,10,14,31]), that would result in production of mature proteins (P1, HC-Pro, P3, 6K1, CI, 6K2, VPg-NIa, NIb, and CP) that also are encoded by other species of the genera Rymovirus, Potyvirus, and "Ipomovirus". Two motifs that are conserved among all potyviruses, the polymerase motif of the NIb protein and the nucleotide binding motif of the CI protein, occur in the WSMV polyprotein sequence at the expected map positions (Fig.…”

“…This initial translation product is proteolytically cleaved into eight mature proteins (Dougherty and Carrington, 1988). At least two of the mature TEV products (Nla and HC-Pro) function as proteinases that catalyze most of the polyprotein processing (Carrington and Dougherty, 1987;Carrington et al, 1989). At least three potyviral proteins (CI, Nla, Nlb) replicase complex (Domier et al, 1987;Dougherty and Carrington, 1988).…”

“…Secondly, contrary to the assumption we made during our search, the cleavage sites of GCMV might not be colinear with those of other picorna-like viruses. Thirdly, another viral protein could act as a protease with a different specificity, as reported in picomaviruses and recently also in potyviruses (51).…”
