A role for proline synthesis and transport in Listeria monocytogenes barotolerance

Abstract: Aims:  To assess the contribution of proline biosynthesis to listerial barotolerance. Methods and Results:  Using a Listeria monocytogenes proBA deletion mutant, incapable of synthesizing proline, together with a proline‐overproducing strain, the contribution of proline synthesis to listerial barotolerance was determined. The ΔproBA strain does not survive as well as the wild type when subjected to treatment of 500 MPa in rich media and 400 MPa in minimal media (c. 1 log lower survival in both conditions). Bet… Show more

“…Also, elevated salt concentrations in a food product may induce uptake of compatible solutes, which in turn stabilize cells during HPP (Abe, 2007). In line with this, a mutant deficient in synthesis of the compatible solute proline showed increased sensitivity to HPP (Considine et al, 2011) The effect of HPP on L. monocytogenes was investigated on the global transcriptomic level by microarray analysis with subsequent RT-PCR on some target genes (Bowman et al, 2008). This indicated that mRNA levels were reduced globally with increasing intensity and duration of the treatment.…”
