A Role for Neutral Sphingomyelinase-mediated Ceramide Production in T Cell Receptor–induced Apoptosis and Mitogen-activated Protein Kinase–mediated Signal Transduction

Abstract: Studying apoptosis induced by T cell receptor (TCR) cross-linking in the T cell hybridoma, 3DO, we found both neutral sphingomyelinase activation and production of ceramide upon receptor engagement. Pharmacological inhibition of ceramide production by the fungal toxin, fumonisin B1, impaired TCR-induced interleukin (IL)-2 production and programmed cell death. Addition of either exogenous ceramide or bacterial sphingomyelinase reconstituted both responses. Moreover, specific inactivation of neutral sphingomyeli… Show more

“…This finding has been of particular interest in regard to T-cell receptor apoptosis pathways. 42 Chi et al investigated the expression of ceramide content in patients with healthy laryngeal mucosa, leukoplakia and laryngeal carcinoma in retrospective review of 178 patients with leukoplakia of the larynx. They found the expression of ceramide to be decreased within tissue samples of leukoplakia and even more in laryngeal carcinoma.…”

“…indeed several mediators known to increase the intracellular levels of ceramide are involved in the early immune response, i.e., TCR triggering, IL-1␤, TNF-␣, IFN-␥, and CD28 ligation (10,(12)(13)(14)(15)(32)(33)(34). When the T cell forms Ag-specific conjugates with APC, a supramolecular activation cluster is observed at the T-APC contact zone.…”
