A rhodamine-benzimidazole based sensor for selective imaging of acidic pH

Abstract: N-(Rhodamine B)-benzimidazole (RB-IM), featuring an intramolecular spiro-benzimidazole, was developed for selective sensing of protons via opening of the spiro-ring to give fluorescent and colored species. The utility of RB-IM was demonstrated by imaging of lysosomes in live cells and staining of the intestine of zebrafish.Scheme 1 pH sensing by RB-IM and rhodamine-lactams.

“…RhB-EDA is a Rhodamine lactam and, as such, is a part of a group of non-fluorescent and colourless compounds marked by intra-molecular spirolactams, which are poised to form highly fluorescent rhodamine derivatives under appropriate conditions via ring opening of the spirolactam [ 35 , 36 , 37 ]. Upon the addition of H + ions ( Scheme 2 ), its structure undergoes a change from the spirolactam to an open ring amide, resulting in a magenta-coloured, highly fluorescent compound [ 35 , 38 ]. The fluorescence spectra showed that RhB-EDA fluorescence is the highest at pH 4.…”

“…A good reversibility of the probe toward pH enabled us to propose a mechanism for the equilibrium of probe REI between pH = 4.2 and pH = 7.2 (Scheme 1). A possible interaction between probe REI and H + is also suitable for the similar phenomenon of other pH probes with the same functional fluorophore as probe REI [39,59,65,66].…”
