A Rapid Detection Method of Brucella with Quantum Dots and Magnetic Beads Conjugated with Different Polyclonal Antibodies

Abstract: Brucella spp. are facultative intracellular bacteria that cause zoonotic disease of brucellosis worldwide. Traditional methods for detection of Brucella spp. take 48–72 h that does not meet the need of rapid detection. Herein, a new rapid detection method of Brucella was developed based on polyclonal antibody-conjugating quantum dots and antibody-modified magnetic beads. First, polyclonal antibodies IgG and IgY were prepared and then the antibody conjugated with quantum dots (QDs) and immunomagnetic beads (IMB… Show more

“…Sun et al detected S. pullorum by polyclonal antibody and blue-silica nanoparticles ranging from 4.4 Â 10 2 to 4.4 Â 10 7 cfu mL À1 [14], and also detected S. pullorum and S. gallinarum by silica and magnetic nanoparticles in milk powder 8.8 Â 10 2 cfu mL À1 [16]. Song et al detected Brucella by using polyclonal antibody-conjugating quantum dots and antibody modified magnetic beads, 10 2 -10 5 cfu mL À1 [28]. Wu et al determined B. melitensis by sensitivity method and using gold nanoparticles-screen-printed carbon electrode 1 Â 10 4 to 4 Â 10 5 cfu mL À1 [29].…”

“…QD immunochromatographic test strips have strict requirements for the immunogenicity of labeling and coating proteins. Dan et al developed a detection of Brucella with QDs and magnetic beads conjugating with different polyclonal antibodies and this method takes 105 min with a detection limit of 10 3 CFU/mL [43]. The method we established only need 10-15 min to obtain nal results with a limit of 1.05 ng/mL.…”
