Arsenic bioavailability to rice plant in paddy soil: influence of microbial sulfate reduction

Purpose High arsenic (As) mobility in anaerobic paddy soil due to microbial Fe-and As-reducing processes results in As accumulation into rice plants. Sulfur (S) also undergoes microbial reducing processes in the anaerobic paddy soil, while this process interacts with the Fe-and As-reducing processes, forms secondary minerals, and thus influences As mobility in paddy soil. This work was carried out to investigate the role of sulfur and sulfate-reducing bacteria (SRB) in As redox transformation and bioavailability to rice plant in an anaerobic paddy soil. Materials and methods Anaerobic incubation experiments with or without sulfate (SO 4 2− ) and SRB were carried out to monitor S and iron (Fe) dynamics and their relations to As speciation and release to soil solution. Rice cultivation in pot experiment was then carried out to check the SO 4 2− amendment in bioavailability and uptake into rice plants.Results and discussion The inoculation of an enriched SRB community into the sterilized paddy soil increased reduction and release of As to the soil solution after flooding, showing its ability in arsenate (As(V)) as well as SO 4 2− reduction to arsenite (As(III)) and sulfide (S 2− ), respectively. Sulfate addition (2 and 100 mM) into the anaerobic paddy soil increased the reduction of SO 4 2− and ferric iron (Fe 3+ ) and resulted to lower dissolved As in the soil solution, which limited As mobility in the paddy soil. Application of SO 4 2− (100 mg kg −1 ) into paddy soil finally decreased the concentration of dissolved As in the soil solution by 23.5 % and also decreased As content in rice roots and iron plaque by 22.6 and 30.5 %, respectively. Conclusions The results revealed the important role of sulfur and the SRB activity in As speciation and mobility in anaerobic paddy soil, implying a lower As bioavailability to rice plants under sulfur-enriched anaerobic paddy soil, and an efficient way to reduce As accumulation in rice plants by sulfur fertilization in paddy soils.

To follow the anaerobic degradation of organic matter in tidal-flat sediments, a stimulation experiment with 13 C-labeled Spirulina biomass (130 mg per 21 g sediment slurry) was conducted over a period of 24 days. A combination of microcalorimetry to record process kinetics, chemical analyses of fermentation products and RNA-based stable-isotope probing (SIP) to follow community changes was applied. Different degradation phases could be identified by microcalorimetry: Within 2 days, heat output reached its maximum (55 lW), while primary fermentation products were formed (in lmol) as follows: acetate 440, ethanol 195, butyrate 128, propionate 112, H 2 127 and smaller amounts of valerate, propanol and butanol. Sulfate was depleted within 7 days. Thereafter, methanogenesis was observed and secondary fermentation proceeded. H 2 and alcohols disappeared completely, whereas fatty acids decreased in concentration. Three main degraders were identified by RNA-based SIP and denaturant gradient gel electrophoresis. After 12 h, two phylotypes clearly enriched in 13 C: (i) Psychrilyobacter atlanticus, a fermenter known to produce hydrogen and acetate and (ii) bacteria distantly related to Propionigenium. A Cytophaga-related bacterium was highly abundant after day 3. Sulfate reduction appeared to be performed by incompletely oxidizing species, as only sulfate-reducing bacteria related to Desulfovibrio were labeled as long as sulfate was available.

Under seasonally fluctuating redox conditions in sediment of Kiel Bay (eastern Baltic Sea), viable counts (MPN) of sulfate-reducing bacteria (SRB) ranged between 4 X 102 and 7 X 104 cm-3 These MPN appeared fairly independent of ambient redox potentials and followed peaks of phytoplankton productivity in the water column with a time lag of 2 to 3 wk. The relative proportions of SRB using acetate, lactate or succinate as their electron donors fluctuated widely. Shells of the clam Arctica islandfca, which can survive anoxia, were, even In oxlc sediments, colonized by eplzolc SRB Significant differences between the abundance of epizoic SRB and SRB from ambient sediment were not detected.In terms of enrichment kinetics, however, epizo~c SRB, and particularly those d e p e n d~n g on succinate as electron donor, showed quicker responses. It 1s hypothesized that SRB associated with benthic infauna represent the biogeochemically more reactive group.
