A Quantitative Proteomic Approach to Identify Significantly Altered Protein Networks in the Serum of Patients with Lymphangioleiomyomatosis (LAM)

Abstract: Lymphangioleiomyomatosis (LAM) is a rare and progressive cystic lung condition affecting approximately 3.4–7.5/million women, with an average lag time between symptom onset and diagnosis of upwards of 4 years. The aim of this work was to identify altered proteins in LAM serum which may be potential biomarkers of disease. Serum from LAM patient volunteers and healthy control volunteers were pooled and analysis carried out using quantitative 4-plex iTRAQ technology. Differentially expressed proteins were validat… Show more

“…The level of Prostatic Specific Antigen (PSA) was determined by human Abcam’s Prostate Specific Antigen PSA ELISA kit or also commonly termed tissue Kallikrein-3 (KLK3) [ 16 ]. Interferon γ (IFN-γ) was determined by ELISA quantitative sandwich immunoassay kit provided from the Eagle Biosciences, Korea [ 17 ].…”

“…However, there are currently no biomarkers for the reliable screening of patients with ccRCC [8][10]. Human serum contains a complex array of peptides, and some of these could serve as biomarkers because their presence/absence or relative abundance may be correlated with certain diseases and could thus be useful for prognosis or diagnosis [11][13].…”

“…The identification of biomarkers for the early detection of cancer could lead to the development of efficient treatments, reduce suffering, and lower mortality rates [7] . “…In all three of these studies a MALDI/TOF/TOF approach was used with iTRAQ labeling. Banville et al used ESI with iTRAQ to search for serum biomarkers that could potentially distinguish lymphangioleiomyomatosis from other respiratory conditions …”
