RESUMO:As sementes de guaraná (Paullinia cupana Kunth, Sapindaceae) são utilizadas como estimulante e os principais constituintes químicos são as metilxantinas e os taninos, tais como catequina e epicatequina. Este trabalho teve por objetivo investigar um método por cromatografia líquida de alta eficiência (CLAE) para quantificar, simultaneamente, cafeína, teofilina, teobromina, catequina e epicatequina em pó de guaraná. Os valores obtidos foram comparados com os resultados encontrados no método espectrofotométrico da Farmacopéia Brasileira IV ed. Pelo método cromatográfico, os teores de metilxantinas e taninos totais foram de 4,04% e 1,48%, respectivamente. Os teores de metilxantinas e taninos totais, obtidos pelo método espectrofotométrico, foram de 4,88% e 4,05%, respectivamente. Acredita-se que, nas condições experimentais empregadas, o método espectrofotométrico não foi específico, quantificando outras substâncias fenólicas presentes no guaraná. Portanto, sugere-se o emprego desse método cromatográfico no controle de qualidade do guaraná em pó, por ser preciso, sensível, específico e rápido. Unitermos:Controle de qualidade, planta medicinal, guaraná, cafeína, catequina, CLAE.ABSTRACT: "Determination of tannins and methylxanthines in powdered guarana (Paullinia cupana Kunth, Sapindaceae) by high performance liquid chromatography". The seeds of Paullinia cupana Kunth, Sapindaceae, are used as a stimulant and the main chemical constituents are the methylxanthines and tannins, as catechin and epicatechin. This study aimed to investigate a method by high performance liquid chromatography (HPLC) to quantify both caffeine, theophylline, theobromine, catechin and epicatechin in powdered of P. cupana. The values obtained were compared with the results of the spectrophotometric method of the Brazilian Pharmacopoeia IV ed. For the chromatographic method, the levels of methylxanthines and total tannins were 4.04% and 1.48%, respectively. The levels of methylxanthines and total tannins obtained by the spectrophotometric method, were 4.88% and 4.05%, respectively. It is believed that, under our experimental conditions, the spectrophotometric method was not specific and quantified others phenolic substances present in the P. cupana. Therefore we suggest the use of this chromatographic method in quality control of P. cupana powder, to be accurate, sensitive, specific and fast. Tanacetum parthenium (feverfew) is an herb that is commercialized worldwide as a therapeutic treatment for migraine. Its pharmacological effect is mainly due to the presence of the sesquiterpene lactone parthenolide as well as of flavonoids. So far, there are no studies on standardization of pre-formulations or phytomedicines containing this herb. The present study aimed at developing a pre-formulation using a standardized spray-dried extract of feverfew and further designing and standardizing enteric coated tablets. In this work, the spray-dried extract of feverfew was evaluated for its parthenolide, santin and total flavonoid content, parthenolide solubility, particle size, tapped density, hygroscopicity, angle of repose and moisture content. Tablets containing the spray-dried extract were tested for their average weight, friability, hardness, and disintegration time. The total flavonoid and parthenolide contents in the spray-dried extract were 1.31 % and 0.76% w/w, respectively. The spray-dried extract presented consistent pharmacotechnical properties and allowed its tableting by direct compression. Tablet properties were in accordance with the proposed specifications. The procedures described herein can be used to prepare and evaluate pre-formulations of feverfew with adequate properties for the development of a high-quality phytomedicine.Uniterms: Enteric coated tablets. Seu efeito farmacológico é creditado principalmente à lactona sesquiterpênica partenolídeo e flavonóides. Até o momento não existem estudos sobre a padronização de pré-formulações ou o desenvolvimento de fitoterápicos com tanaceto. Logo, o objetivo deste trabalho foi obter comprimidos de revestimento entérico a partir de extrato seco e padronizado de tanaceto. Neste trabalho, o extrato seco do tanaceto obtido pelo método de spray drying foi avaliado quanto ao teor de partenolídeo, presença da santina, teor de flavonóides totais, solubilidade do partenolídeo, tamanho de partícula, ângulo de repouso, densidade, análise higroscópica e teor de umidade. A partir do extrato seco obtiveram-se comprimidos que foram revestidos em leito de jorro. Os comprimidos revestidos foram avaliados com relação ao peso médio, friabilidade, dureza e desintegração. O teor de flavonóides totais e de partenolídeo no extrato seco foram 1,31% e 0,76% (p/p), respectivamente. O extrato seco apresentou características farmacotécnicas satisfatórias permitindo a obtenção de comprimidos pelo método de compressão direta. As propriedades dos comprimidos revestidos estão de acordo com as especificações da literatura. Os procedimentos utilizados nesse trabalho podem ser utilizados para obter extrato seco e fitoterápicos de T. parthenium com alto padrão de qualidade.

A validated capillary gas chromatography method for guaco (Mikania glomerata S.) quality control and rastreability: from plant biomass to phytomedicines ABSTRACT: This work describes a full validation of a capillary gas chromatography analytical methodology using internal standardization for the quantification of coumarin (1,2-benzopyrone) in guaco (Mikania glomerata Spreng -Asteraceae) products: syrup, plant and its extract, including the stability study of the phytomedicine. For the analysis, it was used an HP-5 capillary column (30 m x 0.32 mm x 0.25 µm), hydrogen at a flow rate of 1.8 mL/min and the increasing temperature gradient was: 100 °C to 250 °C, 15 ºC/min. The temperature of injector (split 1:20) and detector were kept at 250 °C and 270 °C, respectively. The retention times of the standards for the above conditions were 2.86 minutes for 1, 2, 3, 4-tetramethylbenzene, 4.45 min for piperonal (internal standards), and 5.36 minutes for coumarin. After extraction procedure, the recovery of coumarin determined for plant raw material was 101.6 %, while for syrup it was 100.8 %. Detection and quantification limits were 0.5 µg/mL and 1.5 µg/mL, respectively. Precision was determined for all samples and the results were lower than 2.5 %. The total amount of coumarin in plant raw material, its extract and syrup were 0.38 % w/w, 1.33 mg/mL and 0.143 mg/mL, respectively.

RESUMO:Albizia lebbeck (L.) Benth. é popularmente conhecida no Brasil por coração-de-negro. É uma espécie aclimatada nesse país. As sementes dessa espécie são utilizadas no tratamento de diarréia, disenteria e hemorróidas e as folhas, no tratamento de úlceras e nas conjuntivites alérgicas. Este trabalho foi realizado com a finalidade de determinar as características macroscópicas e microscópicas das folhas e sementes dessa planta, uma vez que estas são as partes usadas popularmente. Algumas características de valor na sua diagnose foram apontadas e ilustradas por fotomicrografias. Reações de fitoquímica e a determinação do conteúdo de água e de cinzas foram realizadas no pó das folhas e sementes de A. lebbeck. Saponinas, alcalóides e cumarinas foram detectados nas folhas e sementes; flavonóides foram também identificados nas folhas. O teor de umidade foi 6,78% e 7,91%, respectivamente, para o pó das folhas e sementes. Os valores de cinzas totais e cinzas insolúveis em ácido foram 9,60% e 0,62% para as folhas e 4,13% e 0,21% para as sementes. Os dados obtidos são importantes para o controle de qualidade dessa planta medicinal. O extrato bruto da planta não apresentou atividade antibacteriana.Unitermos: Albizia lebbeck, coração-de-negro, morfoanatomia, planta medicinal.ABSTRACT: "Pharmacognostic characterization of leaves and seeds of Albizia lebbeck (L.) Benth. (Fabaceae)". Albizia lebbeck (L.) Benth., is popularly known in Brazil as "coração-de-negro". It is a species acclimatized in Brazil. The seeds of this species are used to treat diarrhea, dysentery and hemorrhoids and the leaves are used for the treatment of ulcers and allergic conjunctivitis. The purpose of this work was to determine the macroscopic and microscopic characteristics of the leaves and seeds of this plant, since these are the parts used popularly. Some characters of diagnostical value are pointed out and illustrated by photomicrographs. Phytochemistry reactions and determination of water and ash were done on the powder of A. lebbeck. Saponins, alkaloids, coumarins were detected in the leaves and seeds; flavonoids were also detected in the leaves. The content of water was 6.78% and 7.91%, respectively, for leaf and seed powder. The values of total ashes and acid insoluble ashes were 9.60% and 0.62% for leaf, and 4.13% and 0.21%, for seed powder. The data obtained are important for the quality control of this medicinal plant. The crude extract of this plant did not show antibacterial activity.
