A possible contribution of mRNA secondary structure to translation initiation efficiency inLactococcus lactis

Abstract: Gene expression signals derived from Lactococcus lactis were linked to lacZ‐fused genes with different 5′‐nucleotide sequences. Computer predictions of mRNA secondary structure were combined with lacZ expression studies to direct base‐substitutions that could possibly influence gene expression. Mutations were made such that the DNA sequence upstream of the ATG start codon was not changed. Moreover, care was taken that the substitutions, which were all within the first six codons, neither affected the amino aci… Show more

“…Van de Guchte et al . () showed through of in silico analysis that the mRNA secondary structure might contribute to improve the initial efficiency of translation. The characterization of the L. lactis translatome allowed determinate the proportion of mRNA involved in translation, as well as its ribosome occupancy and density (Picard et al ., ).…”

“…For many RNA molecules, the right secondary structure is a prerequisite for their function. This is well known for secondary structure elements of messenger RNAs, and for the genomes of RNA viruses [38–46]. It means two things.…”

“…So RBS and start codon should play major role in translation efficiency. In addition, negative effect on translation initiation of secondary structure that sequestered RBS was reported in L. lactis [53].…”
