A polysaccharide of PFP-1 from Pleurotus geesteranus attenuates alcoholic liver diseases via Nrf2 and NF-κB signaling pathways

Abstract: A polysaccharide named as PFP-1 was isolated from Pleurotus geesteranus fruiting body, and the potential investigations on ameliorating oxidative stress and liver injury against alcoholic liver disease (ALD) were processed...

“…Alcohol abuse may first result in excessive accumulation of acetaldehyde, a highly toxic metabolite that produces excessive active oxygen radicals and increases oxidative stress damage [ 2 ]. Besides, abnormalities in the metabolic pathway of alcohol dehydrogenase can generate a variety of harmful free radicals that attack lipids and cell membrane proteins, resulting in liver oxidative stress and lipid peroxidation [ 3 ]. Drug therapy is considered the current primary treatment for alcoholic liver injury, while most therapeutic drugs still have a series of unpleasant side effects.…”

“…Our previous reports and other reports have proved that bioactivities of polysaccharides are strongly associated with the structures. 16,27 So, we processed the depth characterizations of PGPs. Referencing our animal results and other reported literature, we can deduce that the repeating units of →4)-α-D-Glcp-(1→ in the main chains and the →4)-β-D-Glcp-(1→ as the branched chains at O6 site, accompanied with proper molecular weights and partially reticulated properties of PGPs contributes to its the potential hepatic and intestinal protection.…”

“…The P. geesteranus strain used in the present study was preserved by our Laboratory (Taian, China), and the PGPs were obtained according to previous methods. 16 Briefly, the mycelium of P. geesteranus was delipidificated by refluxing with ethanol (95%, 4 h, 60 °C) with a solid-liquid ratio of 1 : 20 (m : v, g mL −1 ), and then extracted three times with distilled water at 90 °C for 5 h. After centrifugation (3000 rpm, 15 min), the supernatant was mixed with ethanol (1 : 3, 95%, v/v), and the resulting precipitate was considered as PGPs followed by deproteination (Sevage method), dialysis and lyophilisation, respectively.…”
