A Palm Germ-Radar (PaGeR) for rapid and simple COVID-19 detection by reverse transcription loop-mediated isothermal amplification (RT-LAMP)

Abstract: The current COVID-19 pandemic caused by SARS-CoV-2 is raging, seriously threatening people's lives. The establishment of rapid and accurate pathogen detection technology is not only critical in this epidemic, but also a reminder that we must always be prepared for possible future outbreaks. Therefore, we developed a Palm Germ-Radar (PaGeR) device for rapid and simple detection of COVID-19 from extracted patient sample RNA by RT-LAMP. The whole procedure of rapid COVID-19 detection is based on 4 simple steps: i… Show more

“…For instance, the IoT-based detection of COVID-19 using LAMP technology has been demonstrated with satisfactory performance [ 81 ]. Similarly, another field-deployable RT-LAMP-based device for onsite virus inactivation and detection was also reported [ 82 ]. These advanced approaches can be extended to MPXV diagnostics.…”

“…The outbreak has therefore begun to enable diagnostic assays for screening and preventing the spread of the disease. Current diagnostic methods have solely relied on the nucleic acid amplification technologies, such as real time - polymerase chain reaction (RT-PCR) [ [4] , [5] , [6] ] and Loop mediated isothermal amplification (LAMP) [ [7] , [8] , [9] ]. Nevertheless, these methods have merits, such as they require expensive instruments and sophisticated operation steps which are the key bottlenecks in testing that dramatically increases cost and assay time from sample-to-answer.…”

“…The device could detect SARS-CoV-2 in 25 min with a limit of detection of 10 5 copies in 25 μL sample. Based on RT-LAMP, the Palm Germ-Radar (PaGeR) platform can detect the COVID-19 viral genome down to 1-5 ×10 3 copies/mL within 1 h with a naked-eye using three detection methods, which were colorimetric, fluorimetric, and lateral dipstick [ 12 ]. However, the platform could not enable real-time in situ quantitative monitoring.…”
