A Novel Drug Design Strategy: An Inspiration from Encaging Tumor by Metallofullerenol Gd@C82(OH)22

Abstract: Cancer remains a major threat to human health worldwide. Cytotoxicity has imposed restrictions on the conventional cytotoxic drug-based chemotherapy. The rapidly-developing nanomedicine has shown great promise in revolutionizing chemotherapy with improved efficiency and reduced toxicity. Gd@C82(OH)22, a novel endohedral metallofullerenol, was first reported by our research group to suppress tumor growth and metastasis efficiently without obvious toxicity. Gd@C82(OH)22 imprisons tumors by facilitating the forma… Show more

“…These results are also considered against a background of the anticancer activity of other metal 8-quinolinolates [174]. A completely new approach to chemotherapy has recently been reported in which a metallofullerenol, Gd@C 82 (OH) 22 , was used to encage rather than kill a tumour [175]. It will be interesting to see how this method develops.…”

“…The biological effects of fullerenols offer a vast field of study with a variety of approaches [5,[27][28][29]. Therapeutic effects of fullerenols [5,14,30] are usually explained by their radical scavenging properties.…”

“…The possibility of fullerenes acting as mitochondria protonophores allows them to be considered anti-aging antioxidants [ 15 , 16 ]. Hydroxylated Gd@C 82 has antineoplastic activity simultaneously with low toxicity [ 17 , 18 ].…”
