A Novel DNA Biosensor Using a Ferrocenyl Intercalator Applied to the Potential Detection of Human Population Biomarkers in Wastewater

Abstract: A new label-free electrochemical DNA (E-DNA) biosensor using a custom synthesized ferrocenyl (Fc) double-stranded DNA intercalator as a redox marker is presented. Single-stranded DNA (ssDNA) was co-immobilized on gold electrodes with 6-mecarpto-hexanol to control the surface density of the ssDNA probe, and hybridized with complementary DNA. The binding of the Fc intercalator to dsDNA was measured by differential pulse voltammetry. This new biosensor was optimized to allow the detection of single base pair mism… Show more

“…The number of endogenous biomarkers excreted by humans that were successfully quantified in wastewater is very limited94041. Furthermore, only one of those focused on an endogenous biomarker as an indicator for the overall health of population (8-iso-PGF 2α )9 while another two studies investigated the use of endogenous biomarkers for estimating population size (5-hydroxyindoleacetic acid40 and DNA41).…”

The development and validation of endogenous biomarkers for the assessment of public health through WBE is indeed complicated since there are a limited number of validated endogenous urinary biomarkers for routine clinical use.…”

“…This data can be achieved with biosensors . Biosensors have already been applied in WBE to screen PSA, prostate cancer biomarker (Yang et al, ), DNA (Yang et al, ), and cocaine (Yang et al, ). There are, however, several issues that need to be addressed; mainly the relatively low sensitivity of biosensors and most probably low selectivity.…”

“…Thus, such devices are of particular interest in the area of personal diagnostics, for example, forp oint-of-care devices and on-site monitoring. [4] An example of al abel-free approach is the use of compounds that are able to intercalate into the DNA double helix. One of the main advantages of using intercalating compounds is the fact that these molecules can interactw ith any given DNA sequence.…”

“…Synthesis of af errocene-labeled intercalating compound for the detection of double-stranded DNA (dsDNA) was recent-ly reported. [4] However, this strategy is limited by the oxidation of the 1e À redox species,t hat is, the ferrocene derivative, which in the case of low dsDNA coverage leads to low currents. In contrast, redoxe nzymes conjugated to intercalation compounds offer much higher efficiency at low dsDNAc overage because of the amplification of the amperometrics ignal based on the catalytic redox conversion of as pecific substrate.…”
