A novel class of small RNAs in mouse spermatogenic cells

Abstract: Small noncoding RNAs, including small interfering RNAs (siRNAs) and micro RNAs (miRNAs) of ∼21 nucleotides (nt) in length, have emerged as potent regulators of gene expression at both transcriptional and post-transcriptional levels in diverse organisms. Here we report the identification of a novel class of small RNAs in the mouse male germline termed piwi-interacting RNAs (piRNAs). piRNAs are ∼30 nt in length. They are expressed during spermatogenesis, mostly in spermatids. piRNAs are associated with MIWI, a s… Show more

“…Yb encodes a novel protein [20], while piwi encodes a highly conserved protein involved in regulation of microRNA (miRNA) production [21,22,[51][52][53][54]. Yb exerts its effect on GSC function by regulating expression of piwi and hh in TFs and cap cells [19].…”

“…Targeted disruptions of the two genes resulted in decreased DNA methylation in retrotransposon sequences and increased levels of their RNAs (Aravin et al 2007b(Aravin et al , 2008Kuramochi-Miyagawa et al 2008). In postnatal testes, meiotic spermatocytes and round spermatids express piRNAs of 25-32 nt in length, which are called pachytene piRNAs (Aravin et al 2006;Girard et al 2006;Grivna et al 2006a;Lau et al 2006;Watanabe et al 2006). They interact with PIWIL2 or PIWIL1 (MIWI) (Kuramochi-Miyagawa et al 2001;Deng and Lin 2002) and have diverse sequences that are mapped in clusters in unannotated regions.…”

“…PIWIL1 is localized to perinuclear structures called chromatoid bodies (Grivna et al 2006b;Kotaja et al 2006;Aravin et al 2009;Chuma et al 2009), which are the presumed sites of PTGS, analogous to the processing bodies in somatic cells (Liu et al 2005). However, since only 17%-20% of the total pachytene piRNAs corresponds to retrotransposons (Aravin et al 2006;Girard et al 2006;Grivna et al 2006a;Lau et al 2006;Watanabe et al 2006), their role in retrotransposon silencing is less clear. A partial deletion of a pachytene piRNA cluster, however, was reported to result in increased expression of LINE-1 ORF1 proteins (Xu et al 2008).…”

“…The continuing discovery of novel small RNA such as the class characterized by 26-31 nucleotides and containing "piwi-interacting RNAs", or piRNA, implicated in spermatogenesis (Aravin et al, 2006;Girard et al, 2006;Grivna et al, 2006;Lau et al, 2006;Watanabe et al, 2006) will increase the need for improvements in differentiating miRNAs from other small RNAs. In summary, we developed a novel monoclonal antibody 4F9 with the capability of capturing Ago-associated miRNAs in mammalian cells and may help to elucidate their target mRNAs.…”
