A novel circular RNA, hsa_circ_0043278, acts as a potential biomarker and promotes non-small cell lung cancer cell proliferation and migration by regulating miR-520f

Abstract: More and more circular RNAs (circRNAs) revealed to play a critical role in the initiation and progression of cancer, however, the effects of circRNAs on non-small cell lung cancer (NSCLC) remain largely undetermined. In the present study, we screened the dysregulated circRNAs in paired NSCLC and normal samples from GEO database and identified circ_0043278 was to be significantly up-regulated in NSCLC and demonstrated it promotes NSCLC progression in vitro and in vivo. Then, we revealed the expression of miR-52… Show more

“…circ_001569 was identified to serve as a sponge for miR-145, which may prove beneficial as increased miR-145 expression promotes proliferation and invasion in colorectal cancer ( 14 ). Furthermore, circ_0043278 promoted non-small cell lung cancer (NSCLC) proliferation and migration through regulating miR-520 expression ( 15 ). Previous studies have also reported that circ_0016788 regu-lated hepatocellular carcinoma (HCC) tumorigenesis through the miR-486/cyclin-dependent kinase 4 pathway ( 16 - 18 ) and circRNA-mitochondrial tRNA translation optimization 1, whose expression was decreased in HCC, and was observed to sequester miRNA-9 and suppress HCC progression.…”

“…A total of 48 h after transfection, cell invasion was measured using a Transwell invasion assay, as described in a previous study ( 22 ). Wound migration were observed under a light microscope at ×40 magnification and cell invasion were observed under a light microscope at ×200 magnification.…”

“…Recently, circRNAs are being found to function in multiple disorders progression (Memczak et al, 2013; Salzman, Chen, Olsen, Wang, & Brown, 2013), such as aging (Knupp & Miura, 2018), osteoarthritis (H. Z. Li, Lin, Xu, Lin, & Lu, 2018; Z. B. Zhou et al, 2019), and cancer (Cui et al, 2019; Fu, Jiang, Li, Hu, & Guo, 2018; X. Huang, Zhang, et al, 2019; Q. Zhong, Huang, Wei, & Wu, 2019). Of note, circRNAs are reported to exert diverse functions in cancer tumorigenesis (S. Meng, Zhou, et al, 2017), metastasis (Shi et al, 2019; Wei et al, 2019) and even chemoresistance (W. Huang, Yang, et al, 2019).…”
