A novel C-terminal heat shock protein 90 inhibitor that overcomes STAT3-Wnt-β-catenin signaling-mediated drug resistance and adverse effects

Abstract: Rationale: The heat shock protein (Hsp) system plays important roles in cancer stem cell (CSC) and non-CSC populations. However, limited efficacy due to drug resistance and toxicity are obstacles to clinical use of Hsp90 inhibitors, suggesting the necessity to develop novel Hsp90 inhibitors overcoming these limitations. Methods: The underlying mechanism of resistance to Hsp90 inhibitors was investigated by colony formation assay, sphere formation assay, western blot analysis,… Show more

“…Taking into consideration the protective role of HSPs, the overexpression of chaperones was reported in various types of solid and hematological malignancies [ 25 , 26 , 27 , 28 , 29 ]. High expression of HSPs also correlated to the chemo- and radioresistance of tumors indicated the possibility of employing these proteins as prognostic and/or diagnostic markers [ 26 , 30 , 31 , 32 , 33 , 34 ].…”

“…Serum was taken from mice after cardiac puncture as stated in a previous report 37 to detect hepatic or renal toxicities following treatment with either chemotherapy (a combination of 5‐FU and Oxa) or anti‐PD‐L1 immunotherapy. Measurements of serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine and blood urea nitrogen (BUN) were made using a Fuji DRI‐Chem 3500s veterinary hematology analyser (Fuji, Tokyo, Japan).…”

“…Heat shock protein 90 (hsp90) inhibitors show better results in clinical use, but in therapy, there is resistance that maybe correlated with CSCs in lung cancer. However, there is a new Hsp90 inhibitor named NCT-80 that could reverse CSCs resulting to resistance by regulating STAT3/Wnt/β-catenin signaling pathways ( 81 ). RNF168, a E3 ubiquitin ligase, is downregulated in lung adenocarcinoma, but upregulated in squamous cell carcinoma; the overexpression of RNF168 could inhibit the CSC features (such as sphere-formation ability, stemness markers ALDH) of NSCLC cells.…”
