A novel apoptosis‐inducing protein from Helicobacter pylori

Abstract: SummaryHelicobacter pylori infection induces apoptosis in gastric epithelial cells. Here, we report a novel apoptosis-inducing protein that functions as a leading factor in H. pylori -mediated apoptosis induction. We purified the protein from H. pylori by separating fractions that showed apoptosis-inducing activity. This protein induced apoptosis of AGS cells in a dosedependent manner. The purified protein consisted of two protein fragments with molecular masses of about 40 and 22 kDa, which combined to consti… Show more

“…6). Although both spots were only weakly antigenic in our immunoblots (11) the protein is known to be a virulence-and apoptosis-inducing factor of H. pylori that occurs in the form of two fragments (25,26). Additionally, it was hypothesized that the protein is membrane-associated (25), and here the first 36 amino acids were not covered in the PMFs so that a cleavage of a signal sequence might have occurred.…”

“…H. felis and H. suis induce apoptosis in the murine gastric epithelial cell line GSM06 and in the human gastric adenocarcinoma cell line AGS (unpublished results). Gamma-glutamyltranspeptidase, which has been associated with the induction of apoptosis in gastric epithelial cells and the inhibition of T-cell proliferation by H. pylori (125,195,201), is also produced by gastric non-H. pylori Helicobacter species (Table 2).…”

“…Ultrathin (80-nm) sections of tissue fixed in phosphatebuffered saline (PBS) containing 2% paraformaldehyde and 2.5% glutaraldehyde were postfixed with 2% osmium tetroxide and embedded in Epon resin. Sections were stained with uranyl acetate and lead acetate and examined using a JEM-1220 electron microscope at 80 kV (Jeol Ltd., Tokyo, Japan) as described previously (57).…”
