A novel and cost effective methodology for qualitative screening of alkalo-thermophilic cellulase free xylano-pectinolytic microorganisms using agricultural wastes

Abstract: Qualitative screening of alkalo-thermophilic cellulase free xylano-pectinolytic microorganisms was done on agricultural residues. Since xylan is an expensive substrate for the isolation of xylanase producing microorganisms, the possibility of using wheat bran for screening of these microorganisms was investigated. Screening was carried out on wheat bran for the selection of xylanolytic microorganisms, on waste paper for the evaluation of cellulase free xylanolytic microorganisms, and on citrus peel for screeni… Show more

“…A concurrent producer of xylanase and pectinase, Bacillus pumilus AJK (MTCC 10414) was isolated from the soil contaminated with the effluents of paper and pulp industry (Kaur et al, 2011). Xylano-pectinolytic enzymes were produced under submerged fermentation in 50 ml of basal medium (g/l: peptone, 5.0; MgSO 4 .7H 2 O, 2.46; pH 7) supplemented with 2% (w/v) wheat bran and 2% (w/v) citrus peel.…”

“…For the qualitative study of 40 isolates with respect to formation of halozone (Plate 1a-c), spot inoculations of the isolates were done on Pectin agar plate, Xylan agar plate following the modified method of Kaur et al (2011) and in case of carboxymethyl cellulase (CMC) agar plate it was according to modified method of Baharuddin et al (2010). Then the plates were incubated for 72 h in Orbital Incubator Shaker (Model CIS-24 BL, Make REMI).…”

“…The use of purified substrates like xylan (Bastien et al 2013;Kamsani et al 2016) and cellulose (Bashir et al 2013) has been done by many workers for qualitative estimation of enzyme activity during screening of xylanase and cellulase producing microbes from termite gut microbiome and has been found to be very expensive. Kaur et al (2011) have replaced the use of highly expensive substrates by agricultural wastes for screening purposes. In view of various industrial applications of cellulase and xylanase enzymes in combination, the present study was aimed at screening and isolation of potential xylano-cellulolytic enzymes producing microbes from termite gut and termitarium using agro-wastes.…”
