A NOMPC-Dependent Membrane-Microtubule Connector Is a Candidate for the Gating Spring in Fly Mechanoreceptors

Abstract: Our results provide strong evidence that the ankyrin-repeat domains of NOMPC structurally contribute to the membrane-microtubule connecting filaments. These filaments, as the most compliant element in the distal tip, are therefore good candidates for the gating springs.

“…TRP channels have been implicated in the mechanotransduction of adult Cho neurons. The TRPN channel NOMPC is important for hearing in adult Drosophila, as well as in larval locomotion and gentle touch behavior (18)(19)(20)(21)(36)(37)(38). Immunostaining revealed expression of NOMPC protein in the cilium of larval Cho neurons (Fig.…”

“…In the class IV subset of these neurons, the piezo channel, which is not a TRP channel, transduces stronger, noxious touch stimuli, while in the class III neurons nompC transduces soft touch. 29) NompC is also essential in the transduction of cuticular deformation by campaniform sensillae, 30) in auditory transduction by Johnston's organ in adult Drosophila 31,32) and in vibration detection by the lateral (pentascolopidial) chordotonal organs in Drosophila larvae. 33) NOMPC is also present in the distal tip of the cilium of leg chordotonal organs, where it is likely to also be involved in sensory transduction for the proprioceptive sense.…”

“…[9][10][11][12] Furthermore, microtubules support channel activity 13 ; for example, the coupling of the transient receptor potential family mechanotransduction channel NompC to an array of microtubules conveys force to gate channel activation. [14][15][16] Dendrite microtubule architecture differs between neuron types 15,[17][18][19][20] in order to support the specific functional requirements of each type. We illustrate that here for the specialized dendrites of class IV nociceptive 21 and class I proprioceptive 22 Drosophila body wall neurons.…”
