A new tospovirus causing chlorotic ringspot on Hippeastrum sp. in China

Abstract: A new tospovirus, HCRV 2007-ZDH, was isolated from a Hippeastrum sp. plant displaying necrotic and chlorotic ringspot symptoms in Yunnan province. This virus isolate was characterized based on particle morphology and RNA sequences analyses. Quasi-spherical, enveloped particles measuring about 70-100 nm, typical of tospoviruses, were observed in sap and cells of the infected plants. Transmission studies by inoculating this isolate mechanically to Hippeastrum sp. confirmed that 2007-ZDH is the causal agent of th… Show more

“…A phylogenetic tree based on the N-protein coding sequences, one of the ICTV (International Committee on Taxonomy of Viruses) demarcation criteria, is displayed in Figure 1 and includes all the proposed and established tospovirus species, with the main geographically based origin for each clade. In Asia, a number of new potential threats to vegetable crops have been recently discovered infecting tomato, sweet pepper (50,110,150), and ornamental crops (36); particularly worrisome is the recent first report of TSWV in India (97). Meanwhile, Iris yellow spot virus (IYSV) belonging to the Euro-Asian clade has been spreading in Europe (5).…”

“…The properties, host range, transmission, virion morphology and antigenic relationships were initially used to classify/identify different tospoviruses including GBNV (Satyanarayana et al 1998), and the classification based on these criteria is very misleading. Recently, the genetic sequences of several tospoviruses were either partially or fully sequenced, and the genetic relatedness were also determined (Torres et al 2012;Dong et al 2013).…”

“…Total RNA was extracted using RNA Extraction kit (May Gene, China) according to the manufacturer's protocol. The extracted RNA was dissolved in DEPC-treated ddH 2 O, and stored at -20°C until further use. The cDNA was synthesized from total RNA isolated from infected watermelon leaves using PrimeScript TM RT Master Mix kit (TaKaRa, Dalian, China) following the manufacturer's instructions.…”
