As part of a revision of the genus Acanthothecis s. lat. (Graphidaceae) in Brazil, an updated world key to the known species of the genus is presented. From Brazil, the following ten new species are described: A. latispora, with single-spored asci, large, muriform ascospores, and norstictic and stictic acids; A. megalospora, with single-spored asci, very large, transversely septate ascospores, and norstictic and protocetraric acids; A. multiseptata, with 8-spored asci, medium-sized and narrow, transversely multiseptate ascospores, and lacking secondary substances except terpenoids; A. norstictica, with 1–2-spored asci, large, muriform ascospores, and norstictic acid; A. oryzoides, with 8-spored asci, medium-sized, transversely septate ascospores, and norstictic acid; A. rimosa, with 2-spored asci, small to medium-sized, muriform ascospores, and stictic acid; A. roseola, with 1–2-spored asci, large, muriform ascospores, and norstictic, stictic and subnorstictic acids; A. saxicola, with 8-spored asci, small, 3-septate ascospores, and norstictic acid; A. subfarinosa, with 8-spored asci, small, 6–8-septate ascospores, and norstictic acid; and A. submuriformis, with 8-spored asci, small, submuriform ascospores, and lichexanthone. In this study we demonstrate the utility of whole genome shotgun (WGS) metagenomics in study organisms with small genomes to improve upon amplicon-based estimates of biodiversity and microbial diversity in environmental samples for the purpose of understanding ecological and evolutionary processes. We generated a database of full-length and near-full-length ribosomal DNA sequence complexes from 273 lichenized fungal species and used this database to facilitate fungal species identification in the southern Appalachian Mountains using low coverage WGS at higher resolution and without the biases of amplicon-based approaches. Using this new database and methods herein developed, we detected between 2.8 and 11 times as many species from lichen fungal propagules by aligning reads from WGS-sequenced environmental samples compared to a traditional amplicon-based approach. We then conducted complete taxonomic diversity inventories of the lichens in each one-hectare plot to assess overlap between standing taxonomic diversity and diversity detected based on propagules present in environmental samples (i.e., the "potential" of diversity). From the environmental samples, we detected 94 species not observed in organism-level sampling in these ecosystems with high confidence using both WGS and amplicon-based methods. This study highlights the utility of WGS sequence-based approaches in detecting hidden species diversity and demonstrates that amplicon-based methods likely miss important components of fungal diversity. We suggest that the adoption of this method will not only improve understanding of biotic constraints on the distributions of biodiversity but will also help to inform important environmental policy.

Highlights from 10+ Years of Lichenological Research in Great Smoky Mountains National Park: Celebrating the United States National Park Service Centennial

Abstract—Great Smoky Mountains National Park is renowned as one of the most biologically diverse tracts of land in North America and is the most visited national park in the United States. The park comprises ∼830 square miles, epitomizes eastern temperate hardwood forests of North America, and serves as a refuge for nearly 20,000 documented species from microbes to plants and mammals. Lichens comprise one particularly diverse group of organisms in the park. In this study, we review data from our 11 years of lichenological research in Great Smoky Mountains National Park. Based on approximately 6,000 new field collections generated, the park checklist now includes 920 species, a 129% increase over estimates made two decades ago. Nearly a quarter of the lichens reported in the park are known from only a single occurence whereas only 7% of the lichens are known from 20 or more occurences. An assessment of commonness/rarity for all 920 species indicates that nearly half of the park's lichens should be considered to be infrequent, rare, or exceptionally rare. We assessed the distributions of all 920 species and found that 54 are endemic to the southeastern United States, 30 are endemic to the southern Appalachians, and eight occur nowhere else than within the confines of the national park. We discuss biogeographical affinities of the park's lichen biota as a whole, delimiting six regional “floristic” connections. Our 11 years of research have resulted in the discovery of several species presumed to be extinct or near-extinct. We make one new combination (Fuscopannaria frullaniae) and describe five species as new to science, each commemorating National Park Service staff instrumental to the completion of the study: Heterodermia langdoniana, Lecanora darlingiae, Lecanora sachsiana, Leprocaulon nicholsiae, and Pertusaria superiana.
