Summary The pathogenesis of equine recurrent uveitis (ERU) is not completely understood. Persistent infection of pathogenic leptospires in the eyes is the probable primary cause. Consistently, we isolated and characterized in this study leptospires from 32.2% of the intraocular samples collected from 501 horses from the following Western European countries: Germany, Switzerland, Austria, The Netherlands, Luxembourg, Great Britain, Italy and Poland. The vast majority of the leptospiral isolates belonged to serogroup Grippotyphosa (78.2%). The other isolates belonged to the Australis group (14.2%), the Sejroe group (3.6%), the Pomona group (2.5%) and the Javanica group (1.5%). Apart from three isolates that likely belong to Leptospira kirschneri, serovar Dadas, all Grippotyphosa group isolates belonged to L. kirschneri, serovar Grippotyphosa. Analysis with monoclonal antibodies revealed that the isolates differed from Moskva V, the reference strain of serovar Grippotyphosa. However, the Grippotyphosa isolates closely resembled serovar Grippotyphosa, strain Duyster, which has been isolated from a Dutch patient. Because of apparent phenotypic and genotypic differences between the strains Moskva V and Duyster, we propose two different types within serovar Grippotyphosa, namely type Moskva and type Duyster. Distribution of type Duyster is restricted to Western Europe. Strikingly, only a few serovars were isolated from the eyes of horses with ERU. A number of other commonly occurring serovars, such as Copenhageni, were not found, whereas the presence of antibodies in blood and eye fluids against such serovars suggests that infection occurs. Therefore, we hypothesize that, while many pathogenic serovars might be able to penetrate the eyes, only few are capable to persist.

BackgroundLeptospirosis has been implicated as a severe and fatal form of disease in Mayotte, a French-administrated territory located in the Comoros archipelago (southwestern Indian Ocean). To date, Leptospira isolates have never been isolated in this endemic region.Methods and FindingsLeptospires were isolated from blood samples from 22 patients with febrile illness during a 17-month period after a PCR-based screening test was positive. Strains were typed using hyper-immune antisera raised against the major Leptospira serogroups: 20 of 22 clinical isolates were assigned to serogroup Mini; the other two strains belonged to serogroups Grippotyphosa and Pyrogenes, respectively. These isolates were further characterized using partial sequencing of 16S rRNA and ligB gene, Multi Locus VNTR Analysis (MLVA), and pulsed field gel electrophoresis (PFGE). Of the 22 isolates, 14 were L. borgpetersenii strains, 7 L. kirschneri strains, and 1, belonging to serogoup Pyrogenes, was L. interrogans. Results of the genotyping methods were consistent. MLVA defined five genotypes, whereas PFGE allowed the recognition of additional subgroups within the genotypes. PFGE fingerprint patterns of clinical strains did not match any of the patterns in the reference strains belonging to the same serogroup, suggesting that the strains were novel serovars.ConclusionsPreliminary PCR screening of blood specimen allowed a high isolation frequency of leptospires among patients with febrile illness. Typing of leptospiral isolates showed that causative agents of leptospirosis in Mayotte have unique molecular features.

Expression and Distribution of Leptospiral Outer Membrane Components during Renal Infection of Hamsters

The outer membrane of pathogenic Leptospira species grown in culture media contains lipopolysaccharide (LPS), a porin (OmpL1), and several lipoproteins, including LipL36 and LipL41. The purpose of this study was to characterize the expression and distribution of these outer membrane antigens during renal infection. Hamsters were challenged with host-derivedLeptospira kirschneri to generate sera which contained antibodies to antigens expressed in vivo. Immunoblotting performed with sera from animals challenged with these host-derived organisms demonstrated reactivity with OmpL1, LipL41, and several other proteins but not with LipL36. Although LipL36 is a prominent outer membrane antigen of cultivated L. kirschneri, its expression also could not be detected in infected hamster kidney tissue by immunohistochemistry, indicating that expression of this protein is down-regulated in vivo. In contrast, LPS, OmpL1, and LipL41 were demonstrated on organisms colonizing the lumen of proximal convoluted renal tubules at both 10 and 28 days postinfection. Tubular epithelial cells around the luminal colonies had fine granular cytoplasmic LPS. When the cellular inflammatory response was present in the renal interstitium at 28 days postinfection, LPS and OmpL1 were also detectable within interstitial phagocytes. These data establish that outer membrane components expressed during infection have roles in the induction and persistence of leptospiral interstitial nephritis.
